四川省阿壩州藏族羌族大骨節(jié)病的環(huán)境—遺傳交互作用研究

【摘要】:目的:通過(guò)比較分析四川省阿壩州藏族和羌族兩個(gè)民族環(huán)境-遺傳交互作用對(duì)大骨節(jié)病發(fā)病的影響的異同點(diǎn),建立環(huán)境-遺傳交互作用回歸模型,定性定量分析模型中環(huán)境因素和遺傳因素。方法:用問(wèn)卷調(diào)查方法收集了環(huán)境因素中的居民生活方式情況,并對(duì)調(diào)查人群采集了血樣進(jìn)行遺傳因素的分析。利用SAS軟件分析了阿壩州自然環(huán)境數(shù)據(jù)與大骨節(jié)病患病率、分度程度之間的關(guān)系,篩選出線性回歸的環(huán)境因素構(gòu)建了回歸模型;基于病例對(duì)照研究方法分析了兩個(gè)族群生活方式之間的差異,通過(guò)logistic回歸模型分析不同因素對(duì)大骨節(jié)病發(fā)病的影響;利用SNP分型檢測(cè)技術(shù),建立了藏族和羌族生物信息樣本數(shù)據(jù)庫(kù);利用SHEsis軟件比較分析兩個(gè)民族遺傳度差異性;最后建立環(huán)境因素與遺傳因素交互作用模型。結(jié)果:環(huán)境因素分析顯示:海拔高度、氣溫、降水量和發(fā)硒含量均與大骨節(jié)病患病率有顯著相關(guān)(P0.05),其中海拔高度和降水量與大骨節(jié)病分度程度之間呈線性回歸(P0.05)。藏族:高年齡組、非改水、偶爾食用蔬菜、偶爾食用肉類、較差的居住條件、父母均患、父親與母親患病分度為Ⅲ度為大骨節(jié)病致病高危因素;女性、相對(duì)高學(xué)歷非務(wù)農(nóng)、相對(duì)高收入政府提供主食可降低大骨節(jié)病發(fā)病。羌族:高年齡組、相對(duì)低收入、父母均患病、父母親患、蠖葹橹虏∫蛩兀幌鄬(duì)高學(xué)歷和政府提供主食為保護(hù)因素。大骨節(jié)病易感基因的SNP分析顯示,高危位點(diǎn)及基因型藏族為Bcl-2基因的rs1801018位點(diǎn)攜帶AG基因型、rs1564483位點(diǎn)中攜帶AG+GG基因型、rs17759659位點(diǎn)中攜帶AG基因型,Fas基因的rs2234767位點(diǎn)攜帶AG+GG基因型、rs1800682位點(diǎn)攜帶CT+TT基因;羌族為Bcl-2位點(diǎn)的rs9565727位點(diǎn)中攜帶AA+AG, caspase3基因的rs6948位點(diǎn)中攜帶CC+CA基因型。環(huán)境-遺傳的交互作用分析顯示:在藏區(qū)非改水與Bcl-2基因的rs1564483位點(diǎn)的交互作用對(duì)大骨節(jié)病發(fā)病具有危險(xiǎn)性;食用新鮮蔬菜頻率低與Fas基因rsl 800682位點(diǎn)的交互作用對(duì)大骨節(jié)病發(fā)病具有危險(xiǎn)性;父親患病嚴(yán)重與Fas基因rs1800682位點(diǎn)的交互作用對(duì)大骨節(jié)病發(fā)病具有危險(xiǎn)性;母親患病嚴(yán)重與Fas基因rs2234767位點(diǎn)的交互作用對(duì)大骨節(jié)病發(fā)病具有危險(xiǎn)性。羌族人群中,高年齡組與caspase3基因的rs6948位點(diǎn)的交互作用對(duì)大骨節(jié)病發(fā)病具有危險(xiǎn)性。結(jié)論:對(duì)大骨節(jié)病發(fā)病影響的環(huán)境因素和遺傳位點(diǎn)在兩個(gè)民族有所不同,藏區(qū)有更多的環(huán)境因子通過(guò)與不同的遺傳因子之間影響著大骨節(jié)病的發(fā)生和發(fā)展,這也解釋了藏區(qū)大骨節(jié)病發(fā)病率和嚴(yán)重程度明顯高于羌區(qū)的原因。
[Abstract]:Objective: to establish a regression model of environmental genetic interaction between Tibetan and Qiang nationalities in Aba Prefecture, Sichuan Province, by comparing the differences and similarities of the environmental genetic interaction between them on the incidence of Kashin-Beck disease (KBD). Methods: the environmental factors and genetic factors in the environmental factors were collected by means of a questionnaire survey. SAS software was used to analyze the relationship between the natural environment data of Aba Prefecture and the prevalence and degree of Kaschin-Beck disease, and the regression model was established by selecting the environmental factors of linear regression. Based on the case-control study, the differences between the two ethnic groups' lifestyle were analyzed, and the influence of different factors on the incidence of Kashin-Beck disease was analyzed by logistic regression model, and the SNP typing technique was used to detect KBD. The biological information sample database of Tibetan and Qiang nationality was established, the difference of heritability between the two peoples was analyzed by SHEsis software, and the interaction model of environmental factors and genetic factors was established. Air temperature, precipitation and hair selenium content were significantly correlated with the prevalence of Kaschin-Beck disease (KBD), in which there was a linear regression between altitude and precipitation and the degree of KBD.
