跑臺運動對阿霉素聯(lián)合醋酸戈舍瑞林誘導骨質疏松大鼠骨密度和骨代謝的影響

【摘要】:目的研究阿霉素(doxorubicin,DOX)和醋酸戈舍瑞林(goserelin acetate,GA)聯(lián)合作用對大鼠骨密度和骨代謝的影響,以及跑臺運動對DOX聯(lián)合GA誘導的大鼠骨質疏松的防治效果。方法 8周齡雌性SD大鼠64只,被隨機分為8組,每組8只:安靜對照組(SED)、DOX干預組(SED+DOX)、GA干預組(SED+GA)、DOX和GA聯(lián)合干預組(SED+DOX+GA)、跑臺運動對照組(EX)、跑臺運動結合DOX干預組(EX+DOX)、跑臺運動結合GA干預組(EX+GA)、跑臺運動結合DOX和GA聯(lián)合干預組(EX+DOX+GA)。藥物和跑臺運動干預周期均為8周,8周后測試所有大鼠左側股骨骨密度和血清骨代謝指標。結果與其相對應的非藥物干預各組相比較,藥物干預各組大鼠骨密度顯著降低、骨形成指標ALP和BGP顯著降低而骨吸收指標Ca2+和TRACP5b顯著升高;與DOX單獨干預組及GA單獨干預組相比較,DOX和GA聯(lián)合干預組大鼠骨密度顯著降低、骨形成指標ALP和BGP顯著降低而骨吸收指標Ca2+和TRACP5b顯著升高;與其相對應的安靜組相比較,跑臺運動各組大鼠骨密度顯著升高、骨形成指標ALP和BGP顯著升高而骨吸收指標Ca2+和TRACP5b顯著降低。結論 DOX和GA單獨或聯(lián)合作用均可導致大鼠骨質疏松癥的發(fā)生,且DOX和GA聯(lián)合作用誘導大鼠骨質疏松的程度顯著大于DOX或GA單獨作用的結果;跑臺運動可以有效降低DOX和GA單獨或聯(lián)合作用誘導的大鼠骨質疏松。
[Abstract]:Objective to study the adriamycin (doxorubicin, DOX) and Gore Sherry Lin (goserelin acetate, GA acetate) combined effects on bone density and bone metabolism in rats, and the control effect of treadmill exercise on DOX combined with GA induced osteoporosis rats. Methods 8 week old female SD 64 rats were randomly divided into 8 group, 8 rats in each group: control group (SED), DOX group (SED+DOX), GA group (SED+GA), DOX and GA treatment group (SED+DOX+GA), treadmill exercise control group (EX), treadmill exercise combined with DOX intervention group (EX+DOX), treadmill exercise intervention combined with GA group (EX+GA), treadmill exercise combined with DOX and GA treatment group (EX+DOX+GA). Drugs and treadmill exercise intervention period was 8 weeks, 8 weeks after the test of all the rats left femur bone mineral density and bone metabolism indexes. Non drug intervention groups compared with the corresponding results, drug intervention groups the bone density was significantly decreased, bone formation As indicators of ALP and BGP significantly decreased and the bone absorption index of Ca2+ and TRACP5b were significantly increased; with DOX alone GA alone intervention group and intervention group compared, DOX and GA combined intervention group rats significantly decreased bone density, bone formation index ALP and BGP significantly decreased and the bone absorption index of Ca2+ and TRACP5b significantly increased; and the relative should the quiet group compared with treadmill exercise rats significantly increased the bone density, bone formation index ALP and BGP were significantly increased and the bone resorption index of Ca2+ and TRACP5b decreased significantly. Conclusion DOX and GA alone or in combination can lead to osteoporosis rats occurred, and the combined effects of DOX and GA induced rats osteoporosis was significantly higher than DOX or GA alone. Results; treadmill exercise can effectively reduce the induction of DOX and GA alone or in combination with the function of rats with osteoporosis.
