【摘要】：目的建立一種糖尿病難愈性創(chuàng)面大鼠模型。方法將34只雌性Wistar大鼠隨機(jī)分為對(duì)照組和DM組各17只,適應(yīng)性飼養(yǎng)1周后分別給予常規(guī)飼料及高脂高糖飼料喂養(yǎng)4周;DM組腹腔注射小劑量鏈脲佐菌素,對(duì)照組腹腔注射等量檸檬酸-檸檬酸鈉緩沖液,4周后于兩組大鼠背部制作直徑約2.5 cm的圓形創(chuàng)面;創(chuàng)面造模后第3、7、14天,觀察兩組大鼠創(chuàng)面愈合情況并計(jì)算創(chuàng)面愈合率,HE染色法觀察創(chuàng)面組織病理變化。結(jié)果 DM組創(chuàng)面愈合率分別為5.21%±0.94%、31.70%±1.21%、62.20%±1.37%,對(duì)照組分別為20.87%±1.39%、49.99%±1.58%、91.60%±1.58%,兩組同時(shí)點(diǎn)比較P均0.05。與對(duì)照組相比,其創(chuàng)面組織中含有較少的炎性細(xì)胞、纖維母細(xì)胞及新生毛細(xì)血管。結(jié)論高脂高糖飼料飼養(yǎng)聯(lián)合小劑量STZ注射制作2型糖尿病模型,維持高血糖4周后采用外科方法制作潰瘍模型,可成功建立糖尿病難愈性創(chuàng)面模型。
[Abstract]:Objective to establish a diabetic rat model of refractory wound. Methods 34 female Wistar rats were randomly divided into control group (n = 17) and DM group (n = 17). After one week of adaptive feeding, small doses of streptozotocin were injected into the abdominal cavity of DM group, which were fed with conventional diet and high fat and high sugar diet for 4 weeks, respectively. The rats in the control group were treated with citric acid-sodium citrate buffer solution for 4 weeks and then made a circular wound about 2.5 cm in diameter on the back of the two groups. Results in DM group, the wound healing rate was 5.21% 鹵0.940.The wound healing rate in DM group was 31.70% 鹵1.21 + 0.21% 鹵1.370.The wound healing rate in DM group was 20.87% 鹵1.399.99% 鹵1.58% 鹵1.58%, respectively, compared with that in control group (P < 0.05). There were less inflammatory cells, fibroblasts and new capillaries in the wound tissue. Conclusion the model of type 2 diabetes was induced by high fat and high glucose diet and low dose STZ injection. After maintaining hyperglycemia for 4 weeks, the model of diabetic refractory wound can be successfully established by surgical method.
