咖啡酸苯乙酯對2型糖尿病大鼠肝臟保護作用的研究

【摘要】:目的探討咖啡酸苯乙酯(CAPE)對2型糖尿病(T2DM)大鼠肝臟的保護作用及其作用機制。方法 24只雄性SD大鼠隨機分為3組,即空白組,損傷組,保護組。高脂高糖飼料喂養(yǎng)聯(lián)合鏈脲佐菌素誘導建立2型糖尿病大鼠模型,建模成功后,保護組大鼠CAPE灌胃一個月,測量血清中總膽固醇(TC)和甘油三酯(TG)的含量,并對肝臟中丙二醛(MDA)、蛋白羰基化(PCO)的含量以及超氧化物歧化酶(SOD)、過氧化氫酶(CAT)的活性進行測定。結(jié)果損傷組血清中總膽固醇(TC)和甘油三酯(TG)明顯上升,說明糖尿病大鼠血脂升高。在損傷組中MDA、PCO含量明顯升高,而保護組MDA、PCO的含量較損傷組減少(P0.05),說明糖尿病能夠增加肝臟的氧化應(yīng)激,誘導氧化損傷,而咖啡酸苯乙酯的保護能夠降低MDA、PCO的含量,減少氧化損傷;而SOD、CAT在損傷組中含量降低,可能是機體出現(xiàn)的一種自我保護反應(yīng),使得機體本身的抗氧化物質(zhì)大量消耗,咖啡酸苯乙酯可以減少SOD、CAT含量的增加,保護作用比較明顯。結(jié)論糖尿病的發(fā)生可能增加機體的氧化應(yīng)激,咖啡酸苯乙酯能夠有效降低其氧化損傷程度,從而起到對糖尿病肝臟的保護作用。
[Abstract]:Objective to investigate the protective effect and mechanism of capeic acid (Cape) on the liver of rats with type 2 diabetes mellitus T2DM.Methods Twenty-four male SD rats were randomly divided into three groups: blank group and injury group. The model of type 2 diabetic rats was established by high fat and high sugar diet plus streptozotocin. After the model was successfully established, the rats in the protective group were fed with CAPE for one month, and the contents of serum total cholesterol (TC) and triglyceride (TG) were measured. The contents of malondialdehyde (MDAA), protein carbonylation (PCOs) and the activities of superoxide dismutase (SOD) and catalase (CAT) in liver were determined. Results the serum total cholesterol (TCC) and triglyceride (TG) in the injured group were significantly increased. The results showed that the content of MDA-PCO in the injured group was significantly higher than that in the injured group, while the content of MDA-PCO in the protective group was lower than that in the injured group, indicating that diabetes could increase the oxidative stress of the liver and induce oxidative damage. However, the protection of phenethyl caffeate can reduce the content of MDA-PCO and reduce the oxidative damage, while the decrease of SODP-cat in the injured group may be a self-protective reaction, which causes the body to consume a lot of antioxidants. Conclusion the occurrence of diabetes may increase oxidative stress, and phenethyl caffeic acid can effectively reduce the degree of oxidative damage. Thus plays a protective role in diabetic liver.
