外源性高糖及脾切除對小鼠血糖的影響及其機(jī)制

【摘要】:在正常機(jī)體,血糖受到神經(jīng)、內(nèi)分泌和肝臟的嚴(yán)格調(diào)控,維持在一個(gè)狹窄的生理范圍內(nèi)。但是,外源性葡萄糖的持續(xù)應(yīng)用仍然可以引起正常機(jī)體糖代謝紊亂。已有的臨床資料和實(shí)驗(yàn)研究已經(jīng)顯示了脾臟與胰島內(nèi)分泌功能的關(guān)系。我們?yōu)榱颂接懗掷m(xù)外源性葡萄糖干預(yù)是否影響胰島β細(xì)胞的內(nèi)分泌功能及脾臟是否也參與了這一過程。運(yùn)用外科手術(shù)的方法切除小鼠全脾。將實(shí)驗(yàn)小鼠分為8組,分別以不含葡萄糖的純水,10%、15%、20%的葡萄糖飲水喂養(yǎng)正常(4組)和脾切除小鼠(4組)3周。測定小鼠空腹血糖和胰島素水平,計(jì)算各組小鼠高血糖發(fā)生率;正常對照和脾切除對照小鼠、高血糖小鼠進(jìn)行胰島素釋放試驗(yàn)、胰島組織免疫組化雙染色、胰腺組織活性氧簇(ROS)測定、胰島β細(xì)胞葡萄糖轉(zhuǎn)運(yùn)體(GLUT2)表達(dá)分析。20%葡萄糖飲水喂養(yǎng)的正常組和脾切除組小鼠有高血糖出現(xiàn),發(fā)生率分別為60%,75%,高血糖小鼠的血糖濃度分別為8.62±1.27,11.75±0.91 mmol/L,兩組間血糖存在統(tǒng)計(jì)學(xué)差異(p0.05);相比于正常對照組和脾切除對照組,高血糖小鼠胰島素釋放能力也顯著降低(p0.05),ROS水平升高、GLUT2表達(dá)水平降低,并且,胰島β細(xì)胞出現(xiàn)凋亡;在脾切除組高血糖小鼠,上述指標(biāo)變化更顯著。以上結(jié)果說明,20%葡萄糖飲水通過增加胰腺組織ROS水平和降低β細(xì)胞GLUT-2表達(dá)引起胰島β細(xì)胞的凋亡,胰島素分泌能力的低下,進(jìn)而導(dǎo)致小鼠高血糖。脾臟對外源性葡萄糖干預(yù)引起的胰島內(nèi)分泌功能降低具有一定的保護(hù)作用。
[Abstract]:In the normal body, blood sugar strictly regulated by nerve, endocrine and liver, within a narrow physiological range. However, the continuous application of exogenous glucose can still cause normal glucose metabolism. The clinical data and the experimental research has shown that the relationship between the spleen and pancreatic endocrine function. In order to endocrine function to explore whether the sustained and spleen exogenous glucose intervention effect of pancreatic beta cells are also involved in this process. Using the method of surgical resection and splenectomy in mice. The mice were divided into 8 groups, respectively in pure water, without glucose 10%, 15%, 20% of normal glucose water feeding (Group 4) and spleen excision of mice (Group 4) for 3 weeks. Fasting blood glucose and insulin levels were measured and calculated the incidence of hyperglycemia mice; normal control and splenectomy in control mice, hyperglycemia mice of insulin Release test of islet tissue immunohistochemical double staining, pancreatic tissue reactive oxygen species (ROS) determination of glucose, islet beta cell transporter (GLUT2) expression analysis of.20% glucose water feeding normal group and splenectomy mice had high blood glucose, the incidence rates were 60%, 75%, the blood glucose concentration hyperglycemia mice respectively. 8.62 + 1.27,11.75 + 0.91 mmol/L, there was significant difference in blood glucose between the two groups (P0.05); compared with the normal control group and splenectomy group, hyperglycemia mice insulin releasing capacity was significantly lower (P0.05), ROS level increased, the expression level of GLUT2 decreased, and islet beta cell apoptosis in splenectomy group; blood glucose in mice, the index change is more significant. These results indicated that drinking 20% glucose by increasing the level of ROS in pancreatic tissue and reduced beta cell GLUT-2 expression induced apoptosis of pancreatic beta cells, insulin secretion ability is low, The spleen has a protective effect on the decrease of the endocrine function of the islets caused by exogenous glucose intervention.
