RNF180啟動(dòng)子區(qū)甲基化在前列腺癌中的作用機(jī)制

【摘要】:目的通過(guò)檢測(cè)RNF180啟動(dòng)子區(qū)甲基化改變來(lái)闡明前列腺癌發(fā)生和演進(jìn)過(guò)程中RNF180表達(dá)的意義。方法分別采用Western blotting、RT-PCR、甲基化特異性PCR(MSP)及亞硫酸氫鹽測(cè)序(BSP)法檢測(cè)人前列腺癌細(xì)胞系(PC3、LNCap和DU145)與人正常前列腺上皮細(xì)胞(RWPE-1)中RNF180的表達(dá)情況;采用免疫組化法檢測(cè)人前列腺癌組織芯片上前列腺癌及癌旁組織中RNF180蛋白的表達(dá)。結(jié)果前列腺癌細(xì)胞中RNF180基因的m RNA及蛋白的表達(dá)量明顯低于正常前列腺上皮細(xì)胞(P0.05),而前列腺癌細(xì)胞中RNF180啟動(dòng)子區(qū)的甲基化水平卻明顯高于正常前列腺上皮細(xì)胞;RNF180蛋白在前列腺癌組織中的表達(dá)量明顯低于相應(yīng)的癌旁組織。結(jié)論 RNF180啟動(dòng)子區(qū)在前列腺癌細(xì)胞中呈高度非完全性甲基化狀態(tài),這也可能是RNF180在癌細(xì)胞和癌組織中表達(dá)下降或沉默的原因之一。
[Abstract]:Objective to investigate the significance of RNF180 expression in the occurrence and evolution of prostate cancer by detecting the methylated changes of RNF180 promoter region. Methods the expression of RNF180 in human prostate cancer cell lines (PC3,LNCap and DU145) and human normal prostate epithelial cells (RWPE-1) was detected by Western blotting,RT-PCR, methylated specific PCR (MSP) and bisulfite sequencing (BSP), respectively. The expression of RNF180 protein in prostate cancer and paracancerous tissues was detected by immunohistochemistry. Results the expression of m RNA and protein of RNF180 gene in prostate cancer cells was significantly lower than that in normal prostate epithelial cells (P 0.05), while the methylated level of RNF180 promoter region in prostate cancer cells was significantly higher than that in normal prostate epithelial cells. The expression of RNF180 protein in prostate cancer tissues was significantly lower than that in corresponding paracancerous tissues. Conclusion the promoter region of RNF180 is highly incomplete methylated in prostate cancer cells, which may be one of the reasons for the decrease or silence of RNF180 expression in cancer cells and cancer tissues.
