缺氧下膀胱癌細(xì)胞黏附能力和侵襲能力的變化

【摘要】:目的:探討缺氧條件下不同分化膀胱癌細(xì)胞5637和T24生物學(xué)特性變化。方法:將不同分化程度的膀胱癌細(xì)胞5637和T24分別置于正常氧濃度條件下(37℃,體積分?jǐn)?shù)5%CO2和體積分?jǐn)?shù)21%O2)和缺氧(37℃、體積分?jǐn)?shù)10%O2、體積分?jǐn)?shù)5%CO2和體積分?jǐn)?shù)85%N2)環(huán)境中培養(yǎng),以MTT法和Boyden侵襲小室法檢測(cè)兩種膀胱癌細(xì)胞在不同環(huán)境下黏附能力和侵襲能力的差異。結(jié)果:隨著培養(yǎng)時(shí)間的延長(zhǎng),缺氧組和對(duì)照組5637和T24細(xì)胞的黏附率逐漸增加,差異有統(tǒng)計(jì)學(xué)意義(P0.001);5637和T24細(xì)胞黏附率在缺氧24 h與對(duì)照組相比差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05),但缺氧48 h和72 h兩種細(xì)胞的黏附能力均高于對(duì)照組(P0.05);缺氧組5637和T24細(xì)胞的穿膜細(xì)胞數(shù)均高于對(duì)照組(P0.001)。結(jié)論:缺氧條件下膀胱癌細(xì)胞的黏附和遷移能力增強(qiáng)。
