輸精管結(jié)扎對大鼠睪丸網(wǎng)的影響:形態(tài)定量研究

【摘要】:目的:研究大鼠輸精管結(jié)扎后睪丸網(wǎng)的組織學(xué)變化,以探討輸精管結(jié)扎致精子發(fā)生損害的機制。方法:12只10-11周齡的正常雄性(Sprague-Dawley,SD)大鼠,行單側(cè)(隨機選擇左側(cè)或者右側(cè))輸精管結(jié)扎,結(jié)扎位置遠(yuǎn)離附睪尾和陰囊。術(shù)后37天,取大鼠雙側(cè)睪丸。沿垂直于睪丸長軸的方向,把每個睪丸都切成6片,每片切成2塊,全部用甲基丙烯酸樹脂包埋,制作切片,進(jìn)行組織學(xué)觀察,并運用體視學(xué)方法定量研究睪丸網(wǎng)的寬度和總體積。結(jié)果:與未結(jié)扎側(cè)相比,12只大鼠結(jié)扎側(cè)睪丸網(wǎng)的寬度顯著增加了66.1%±29.4%(配對t檢驗,P0.05),但睪丸網(wǎng)的總體積沒有變化。其中7只大鼠結(jié)扎側(cè)睪丸的精子發(fā)生未見明顯損害,但另5只大鼠見明顯損害;單獨看這兩個亞組,結(jié)扎側(cè)睪丸網(wǎng)的寬度都增加了64%-67%,盡管這沒有統(tǒng)計學(xué)意義(P0.16)。結(jié)論:大鼠輸精管結(jié)扎會導(dǎo)致睪丸網(wǎng)增寬,提示結(jié)扎后睪丸液可能淤積,生精小管內(nèi)壓可能增加,這支持輸精管結(jié)扎對精子發(fā)生損害的壓力介導(dǎo)機制。
[Abstract]:Aim: to study the histological changes of testicular reticulum after vasectomy in rats and to explore the mechanism of spermatogenesis induced by vas deferens ligation. Methods: 12 normal male (Sprague-Dawley,SD) rats aged 10-11 weeks were treated with unilateral (left or right) vasectomy, located far from the caudal epididymis and scrotum. After 37 days, bilateral testis were taken from rats. Along the direction perpendicular to the long axis of the testis, each testis was cut into 6 pieces, each of which was cut into 2 pieces, all of which were covered with methacrylic resin, and were made into sections and histologically observed. The width and total volume of testicular reticulum were quantitatively studied by stereological method. Results: compared with the unligated side, the width of testicular reticulum in 12 rats increased significantly by 66.1% 鹵29.4% (pairing t test, P0.05), but the total volume of testicular reticulum did not change. There was no significant damage to spermatogenesis in 7 of them, but obvious damage was found in the other 5 rats. Looking at the two subgroups alone, the width of the ligated lateral testicular reticulum increased by 64-67, although there was no statistical significance (P 0. 16). Conclusion: ligation of vas deferens may lead to the enlargement of testicular reticulum, suggesting that testicular fluid may accumulate after ligation, and the pressure of seminiferous tubules may increase, which supports the pressure-mediated mechanism of vasectomy on spermatogenesis.
