Tamm-Horsfall蛋白對(duì)腎結(jié)石形成的作用機(jī)制

【摘要】:目的 Tamm-Horsfall蛋白(THP)在腎結(jié)石形成過程中可能起一定作用。文中旨在通過建立鈣化性納米微粒(CNPs)大鼠腎結(jié)石模型,測(cè)定大鼠尿液中THP的變化、腎組織病理改變及鈣鹽晶體形成情況,對(duì)THP在腎結(jié)石形成過程中的作用機(jī)制進(jìn)行初步研究。方法取泌尿系患者的結(jié)石標(biāo)本,建立CNPs致大鼠腎結(jié)石模型,制成CNPs混懸液。將48只SD大鼠隨機(jī)分為實(shí)驗(yàn)組(注射CNPs混懸液)和對(duì)照組(注射等滲鹽水),于注射后3 h、6 h、12 h、24 h、1 w、2 w、4 w和8 w收集大鼠尿液,ELISA檢測(cè)尿液中THP的含量,取腎組織行HE染色觀察細(xì)胞的病理改變和對(duì)鈣鹽晶體形成情況進(jìn)行評(píng)分。結(jié)果實(shí)驗(yàn)組24h、1w、2w、4w、8w THP含量[(166.03±3.02)、(173.50±1.78)、(174.55±2.05)、(176.54±2.45)、(177.11±1.76)pg/mL]較3 h[(165.89±2.23)pg/mL]明顯增加(P0.05);亦較對(duì)照組24 h、1 w、2 w、4 w、8 w[(157.65±2.22)、(156.54±1.43)、(159.45±3.21)、(158.63±2.98)、(157.33±2.05)pg/mL]明顯升高(P0.05)。與實(shí)驗(yàn)組6 h鈣鹽晶體評(píng)分(1.0分)比較,3、6、12、24 h的鈣鹽晶體評(píng)分(均為2.0分)增加,2w后鈣鹽晶體評(píng)分增加顯著(3.0分),8w評(píng)分最高(6.7分),差異有統(tǒng)計(jì)學(xué)意義(P0.05)。鈣鹽晶體的評(píng)分與THP含量呈正相關(guān)(r=0.843,P0.05)。結(jié)論尿液中的THP可以促進(jìn)鈣鹽晶體的聚集成核,有利于腎結(jié)石的形成。
[Abstract]:Objective Tamm-Horsfall protein (THP) may play a role in the formation of renal calculi. The aim of this study was to establish a rat model of calcified nanoparticles (CNPs), to determine the changes of THP in urine, the pathological changes of renal tissue and the formation of calcium crystals, and to study the mechanism of THP in the formation of renal calculi. Methods Nephrolithiasis model was established in rats with CNPs and the suspension of CNPs was made. Forty-eight Sprague-Dawley rats were randomly divided into two groups: experimental group (injected with CNPs suspension) and control group (injected with isoosmotic saline). THP levels in urine of rats were collected at 3 h, 6 h, 12 h, 24 h, 1 week, 2 weeks, 4 weeks and 8 weeks after injection.
