C3a、C5a受體缺失減輕IgA腎病腎損傷

【摘要】:目的:探討C3a、C5a及其受體在Ig A腎病發(fā)病中作用及潛在機(jī)制。方法:6~8周清潔級雌性BALB/c小鼠28只,陰性對照組、野生型組、C3a受體敲除組、C5a受體敲除組各7只,采用有活性的仙臺病毒滴鼻經(jīng)鼻黏膜感染聯(lián)合尾靜脈注射建立小鼠Ig A腎病模型,測定24 h尿蛋白量、血清尿素氮、血清肌酐;留取腎組織標(biāo)本,通過直接免疫熒光法檢測腎組織Ig A、C3沉積;采用PAS染色光鏡下觀察腎組織病理改變;采用RT-q PCR法檢測腎組織中TNF-α、TGF-β、IL-1β、IL-6、MCP-1mRNA相對表達(dá)量。結(jié)果:15周后野生型組、C3a受體敲除組、C5a受體敲除組(實(shí)驗(yàn)組)24 h尿蛋白量高于陰性對照組,野生型組高于C3a受體敲除組及C5a受體敲除組,差異具有統(tǒng)計學(xué)意義;血清尿素氮及血清肌酐差異無統(tǒng)計學(xué)意義;實(shí)驗(yàn)組小鼠可見腎臟組織病理改變,且野生型組重于C3a受體敲除組及C5a受體敲除組,陰性對照組腎臟組織病理則無明顯異常。實(shí)驗(yàn)組小鼠腎組織TNF-α、TGF-β、IL-1β、IL-6、MCP-1 mRNA相對表達(dá)量高于陰性對照組,同時野生型組高于C5a受體敲除組及C3a受體敲除組,另外IL-1β、IL-6、MCP-1的mRNA相對表達(dá)量C5a受體敲除組高于C3a受體敲除組。結(jié)論:C3a及C5a受體缺失可減輕Ig A腎病腎損傷,并且C3a受體缺失作用更加顯著。
[Abstract]:Objective: to investigate the role and potential mechanism of C _ 3a and C _ 5a receptor in the pathogenesis of IgA nephropathy. Methods 28 BALB / c mice, 28 female BALB / c mice, 8 weeks clean, negative control group and 7 wild type C3a receptor knockout group were used to establish the model of IgA nephropathy by nasal drip of active Sendai virus (Sendai virus) and tail vein injection. Urine protein content, serum urea nitrogen, serum creatinine were measured at 24 h, renal tissue samples were collected and the deposition of IgA C 3 in renal tissue was detected by direct immunofluorescence, and pathological changes of renal tissue were observed under light microscope with pas staining. RT-Q PCR was used to detect the relative expression of TNF- 偽 -TGF- 尾 -IL-1 尾 -IL-6 and MCP-1 mRNA in renal tissue. Results after 15 weeks, the urine protein content of the wild-type C3a receptor knockout group (experimental group) was higher than that of the negative control group, while the wild type group was higher than the C3a receptor knockout group and the C5a receptor knockout group. The difference was statistically significant. There was no significant difference in serum urea nitrogen and serum creatinine. The pathological changes of kidney were observed in the experimental group and the wild-type group were more serious than those in the C3a receptor knockout group and the C5a receptor knockout group, but there was no obvious abnormality in the renal histopathology in the negative control group. The relative expression of MCP-1 mRNA in renal tissue of experimental group was higher than that of negative control group, while that of wild-type group was higher than that of C5a receptor knockout group and C3a receptor knockout group. In addition, the relative mRNA expression of IL-6MCP-1 mRNA in C5a receptor knockout group was higher than that in C3a receptor knockout group. Conclusion the loss of C _ 3a and C _ 5a receptors can attenuate the renal injury of IgA nephropathy, and the deletion of C _ 3a receptor is more significant.
