【摘要】：目的探討皮質(zhì)抑素(cortistatin,CST)在人腎組織中的表達(dá)情況及其在不同分級(jí)IgA腎病腎組織中的表達(dá)變化。方法收集10例腎癌切除術(shù)患者的癌旁正常腎組織,通過逆轉(zhuǎn)錄聚合酶鏈反應(yīng)(RT-PCR)與Western印跡分別檢測人腎組織中CST mRrNA與蛋白的表達(dá)情況,采用免疫組織化學(xué)(IHC)技術(shù)對(duì)CST于腎組織中的表達(dá)進(jìn)行定位。根據(jù)Lee分級(jí)標(biāo)準(zhǔn)將IgA腎病分為3組:Ⅰ~Ⅱ級(jí)為A組、Ⅲ~Ⅳ級(jí)為B組、Ⅴ級(jí)為C組,每組分別收集10例IgA腎病患者的腎穿組織樣本。采用IHC技術(shù),檢測正常腎組織及不同分級(jí)IgA腎病腎組織中CST的表達(dá)變化。對(duì)臨床指標(biāo)進(jìn)行多重線性回歸分析,評(píng)估其對(duì)IgA腎病腎組織內(nèi)CST表達(dá)量的影響。結(jié)果 RT-PCR與Western印跡結(jié)果均表明腎組織內(nèi)表達(dá)CST,IHC結(jié)果顯示CST表達(dá)于腎小管上皮細(xì)胞。IgA腎病中,病理分級(jí)越高,腎小管中CST表達(dá)越多。多重線性回歸分析顯示,病理分級(jí)與腎組織中CST表達(dá)量相關(guān)(r=0.875,尸0.01)。結(jié)論 CST可能參與IgA腎病病理損傷的炎癥反應(yīng),發(fā)揮抗炎保護(hù)作用。
[Abstract]:Objective to investigate the expression of cortiostatin in human renal tissues and its changes in renal tissues with different grades of IgA nephropathy. Reverse transcriptase polymerase chain reaction (RT-PCR) and Western blot were used to detect the expression of CST mRrNA and protein in human kidney.
