【摘要】：目的：研究賽洛多辛對(duì)左氧氟沙星在慢性細(xì)菌性前列腺炎(CBP)大鼠前列腺組織中藥動(dòng)學(xué)的影響。方法：構(gòu)建SD大鼠慢性細(xì)菌性前列腺炎模型96只,并隨機(jī)分成對(duì)照組(單用左氧氟沙星)和實(shí)驗(yàn)組(賽洛多辛和左氧氟沙星聯(lián)合用藥),每組各48只,分別于給藥后0.125、0.25、0.5、1、2、4、8和12 h每個(gè)時(shí)間點(diǎn)處死6只模型大鼠,采集兩組大鼠的前列腺制作前列腺組織勻漿,高效液相色譜(HPLC)測(cè)定組織中左氧氟沙星濃度,使用3p97軟件計(jì)算相關(guān)藥動(dòng)學(xué)參數(shù)。結(jié)果：對(duì)照組和實(shí)驗(yàn)組的藥-時(shí)曲線均符合一室模型。計(jì)算出的藥動(dòng)學(xué)參數(shù)如下：t1/2分別為(4.34±0.25)h和(5.02±0.28)h,tpeak分別為(0.73±0.20)h和(0.86±0.05)h,CL分別為(1.74±0.19)mg/h和(0.64±0.06)mg/h,Cmax分別(3.62±0.3)mg/kg和(8.47±0.51)mg/kg, AUC0-12分別為(21.58±2.00) mg·h/kg和(56.63±4.30) mg·h/kgo實(shí)驗(yàn)組與對(duì)照組比較,兩組在t1/2上的差異有統(tǒng)計(jì)學(xué)意義(P0.05)；兩組在CL,Cmax,AUC0-12上的差異有顯著統(tǒng)計(jì)學(xué)意義(P0.01)；而兩組在tpeak比較上,差異無統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論：賽洛多辛能明顯提高慢性細(xì)菌性前列腺炎模型大鼠前列腺組織中左氧氟沙星的藥物濃度。
[Abstract]:Objective: to study the effect of sellodoxin on prostatic tissue of rats with levofloxacin in chronic bacterial prostatitis (CBP) rats. Methods: 96 SD rats with chronic bacterial prostatitis were established. They were randomly divided into two groups: the control group (levofloxacin alone) and the experimental group (sellodoxin combined with levofloxacin), 48 rats in each group, and 6 model rats were killed at each time point of 0.125 and 0.25 and 12 hours, respectively. The prostate tissue homogenate of two groups of rats was collected and the concentration of levofloxacin was determined by high performance liquid chromatography (HPLC). The pharmacokinetic parameters were calculated by 3p97 software. Results: the pharmacokinetic parameters of the control group and the experimental group were in accordance with the one-compartment model. The pharmacokinetic parameters calculated were as follows: t 1 / 2 = 4.34 鹵0.25 h and 5.02 鹵0.28 h / h respectively: 0.73 鹵0.20 h and 0.86 鹵0.05 h, respectively, and the pharmacokinetic parameters were 1.74 鹵0.19 mg / h and 0.64 鹵0.06 mg / h, respectively. Compared with the control group, the AUC0-12 of the experimental group was 21.58 鹵2.00 mg 路h / kg and 56.63 鹵4.30 mg 路h / kg, respectively, and that of the control group was 8.47 鹵0.51 mg 路kg 路kg 路h / kg, 3.62 鹵0.3 mg / kg and 8.47 鹵0.51g / kg, respectively. The difference in t1 / 2 between the two groups was statistically significant (P0.05A), the difference between the two groups in the CmaxCmaxAUC 0-12 was statistically significant (P0.01A), and the difference between the two groups in tpeak was significantly higher than that in the control group (P < 0.05), but the difference between the two groups was significant (P < 0.05). Conclusion: the concentration of levofloxacin in prostatic tissue of rats with chronic bacterial prostatitis can be increased obviously.
