孕婦全血多金屬暴露與甲狀腺激素的關(guān)系研究

第一部分孕婦全血多金屬暴露與甲狀腺激素的全環(huán)境組關(guān)聯(lián)分析目的:應(yīng)用全環(huán)境組關(guān)聯(lián)分析(environment-wide association study,EWAS)探索孕婦人群多金屬暴露與甲狀腺激素之間的關(guān)聯(lián)性。方法:基于杭州出生隊列研究(Hangzhou Birth Cohort Study,HBCS)基線調(diào)查數(shù)據(jù),將915名符合納入排除標準的孕婦選為研究對象。在孕20-28(中位數(shù)25)周收集孕婦全血,利用電感耦合等離子體-質(zhì)譜法(ICP-MS)檢測11種全血金屬水平。從醫(yī)院信息系統(tǒng)提取同一時期檢測的血清游離三碘甲狀腺原氨酸(FT3)、總?cè)饧谞钕僭彼幔═T3)、游離甲狀腺素(FT4)、總甲狀腺素(TT4)和促甲狀腺激素(TSH)水平。利用多因素校正單金屬線性回歸模型分別分析各種金屬與甲狀腺激素之間的關(guān)聯(lián)性,金屬水平以三分位數(shù)(設(shè)定第一分位數(shù)為參照組)作為自變量納入模型。對單金屬模型的關(guān)聯(lián)性趨勢進行偽發(fā)現(xiàn)率法(false discovery rate,FDR)校正,將仍有統(tǒng)計學(xué)意義的所有金屬放入多金屬模型互相校正。結(jié)果:單金屬模型篩選出砷(As)、硒(Se)、錳(Mn)、鎳(Ni...
