TGF-β1誘導(dǎo)的Postn、α-SMA和Collagen Ⅰ在宮腔粘連內(nèi)膜組織中表達(dá)上調(diào)且與粘連程度相關(guān)

目的:骨膜蛋白(Postn)參與多種纖維化疾病的病理形成,且與促纖維化因子TGF-β1的信號(hào)通路活動(dòng)密切相關(guān),但在宮腔粘連(IUAs)形成過程中的作用機(jī)制尚不清楚。本研究通過檢測不同程度IUAs患者子宮內(nèi)膜組織中Postn與纖維標(biāo)志物(α-SMA,CollagenⅠ)的mRNA和蛋白質(zhì)表達(dá),并從細(xì)胞水平研究TGF-β1作用與三種目標(biāo)蛋白質(zhì)表達(dá),從而探討Postn、α-SMA和CollagenⅠ在IUAs發(fā)病過程中的作用機(jī)制,及與IUAs嚴(yán)重程度間的關(guān)系。方法:收集明確診斷的輕、中和重度IUAs患者內(nèi)膜組織各20例(實(shí)驗(yàn)組),非宮腔粘連者20例(對(duì)照組),利用實(shí)時(shí)熒光定量PCR技術(shù)、Western印跡技術(shù)和免疫組化染色分別檢測Postn、α-SMA和CollagenⅠ在不同粘連程度IUAs患者的子宮內(nèi)膜組織中mRNA和蛋白質(zhì)表達(dá)差異。同時(shí),Western印跡技術(shù)分別檢測原代ESCs經(jīng)TGF-β1溶液不同時(shí)間、不同濃度作用后Postn、α-SMA和CollagenⅠ蛋白表達(dá)情況。結(jié)果:Postn與α-SMA、CollagenⅠ在IUAs患者子宮內(nèi)膜組織中的mRNA和蛋白質(zhì)表達(dá)水平趨勢一致,...
