TET1在子宮內(nèi)膜病變中的表達(dá)及其對(duì)癌細(xì)胞增殖和遷移能力的影響

【摘要】:目的探討不同類型子宮內(nèi)膜病變中TET1的表達(dá)水平和干擾TET1蛋白表達(dá)對(duì)子宮內(nèi)膜癌細(xì)胞系Hec-1a和Ishikawa細(xì)胞生物學(xué)行為的影響。方法免疫組織化學(xué)檢測(cè)組織中TET1的表達(dá);應(yīng)用干擾TET1表達(dá)的siRNA轉(zhuǎn)染Hec-1a和Ishikawa細(xì)胞,以僅加入轉(zhuǎn)染試劑的細(xì)胞作為陰性對(duì)照;Western blotting檢測(cè)轉(zhuǎn)染效率;SRB法檢測(cè)細(xì)胞增殖能力改變;Transwell侵襲試驗(yàn)檢測(cè)細(xì)胞侵襲能力改變;劃痕試驗(yàn)檢測(cè)細(xì)胞遷移能力改變;流式細(xì)胞術(shù)檢測(cè)細(xì)胞周期改變。結(jié)果子宮內(nèi)膜組織中TET1的表達(dá)隨病理類型進(jìn)展而升高;干擾組TET1蛋白表達(dá)水平明顯低于對(duì)照組;干擾TET1表達(dá)后子宮內(nèi)膜癌細(xì)胞增殖、遷移和侵襲能力均受到不同程度抑制,同時(shí)出現(xiàn)G1/S期細(xì)胞周期阻滯。結(jié)論子宮內(nèi)膜癌及子宮內(nèi)膜增生性病變組織中TET1表達(dá)水平明顯高于正常對(duì)照組織;干擾TET1表達(dá)后子宮內(nèi)膜癌細(xì)胞的增殖、遷移和侵襲能力下降,且出現(xiàn)G1/S期細(xì)胞周期阻滯,提示TET1在子宮內(nèi)膜癌的增殖和轉(zhuǎn)移過程中發(fā)揮重要作用。
[Abstract]:Objective to investigate the effects of TET1 expression and interfering TET1 protein expression on the biological behavior of Hec-1a and Ishikawa cells in different types of endometrial lesions. Methods Immunohistochemical method was used to detect the expression of TET1 in tissues, Hec-1a and Ishikawa cells were transfected with siRNA which interfered with TET1 expression,; Western blotting was used as negative control to detect the efficiency of; Western blotting, and SRB assay was used to detect the change of cell proliferation ability. Transwell invasion test was used to detect the change of cell invasion ability, scratch test was used to detect the change of cell migration ability, and flow cytometry was used to detect the change of cell cycle. Results the expression of TET1 in endometrial tissue increased with the progress of pathological type, and the expression level of TET1 protein in interference group was significantly lower than that in control group. After interfering with the expression of TET1, the proliferation, migration and invasion of endometrial cancer cells were inhibited in varying degrees, and the cell cycle arrest of G _ 1 鈮,
