【摘要】：目的探討孕期亞硝酸鹽暴露對海馬損傷的作用。方法利用C57BL/6J小鼠建立孕期亞硝酸鹽暴露模型,分為對照組(生理鹽水)、低劑量亞硝酸鹽組(60mg/kg)和高劑量亞硝酸鹽組(120mg/kg)。收集出生當(dāng)日(P0)、P7、P14及P30各年齡點(diǎn)仔鼠大腦,用于免疫熒光染色、彗星實(shí)驗(yàn)、Western blotting蛋白半定量分析,對海馬損傷進(jìn)行研究。結(jié)果不論是亞硝酸鹽暴露組還是對照組,仔鼠齒狀回增殖的神經(jīng)干細(xì)胞數(shù)目均隨著年齡的增長逐漸減少;在P0、P7、P14和P30年齡點(diǎn),暴露組仔鼠增殖的神經(jīng)干細(xì)胞的數(shù)量明顯比對照組少,且具有劑量依賴性(P0.05,n=96)。為了鑒別這種抑制作用是否具有選擇性,我們對P0仔鼠室管膜下區(qū)神經(jīng)干細(xì)胞的增殖情況進(jìn)行了觀察。結(jié)果發(fā)現(xiàn),亞硝酸鹽暴露組室管膜下區(qū)的神經(jīng)干細(xì)胞增殖較對照組也減少,同樣具有劑量依賴性(P0.05);亞硝酸鹽暴露組仔鼠門區(qū)炎癥損傷細(xì)胞和凋亡細(xì)胞的數(shù)目比對照組多,具有劑量依賴性(P0.05);彗星實(shí)驗(yàn)結(jié)果顯示,亞硝酸鹽暴露組P0仔鼠海馬細(xì)胞的彗尾比對照組長,具有劑量依賴性(P0.01);與對照組P0仔鼠相比,亞硝酸鹽暴露組仔鼠海馬組織內(nèi)Caspase-8和核因子κB(NF-κB)蛋白的表達(dá)量較高(P0.05)。結(jié)論孕期亞硝酸鹽暴露可通過抑制神經(jīng)干細(xì)胞增殖,促進(jìn)細(xì)胞損傷和凋亡而對仔鼠海馬造成損傷。
[Abstract]:Objective to investigate the effect of nitrate exposure during pregnancy on hippocampal injury. Methods the model of nitrite exposure during pregnancy was established in C57BL/6J mice. The mice were divided into control group (normal saline), low dose nitrite group (60mg/kg) and high dose nitrite group (120mg/kg). The brains of neonatal rats at different age points (P0, P7, P14 and P30) were collected for immunofluorescence staining and semi-quantitative analysis of, Western blotting protein in Comet assay to study hippocampal injury. Results the number of neural stem cells proliferated in dentate gyrus decreased gradually with the increase of age in both nitrous exposure group and control group.
