大鼠側腦室注射瘦素對卵巢摘除給予雌激素子宮腔液2-DE圖譜分析

【摘要】:瘦素(leptin)通過動物下丘腦-垂體軸對其生殖活動予以調節(jié),但瘦素通過下丘腦對子宮調節(jié)作用仍不清楚.本研究在大鼠側腦室微量注射瘦素6 h后,采用雙向凝膠電泳(2-DE)結合基質輔助激光解吸電離飛行時間質譜(MALDI-TOF-MS)技術分離、鑒定了卵巢摘除給予雌激素的子宮腔液蛋白質表達譜.我們獲得了分辨率較高、重復性較好的2-DE圖譜;質譜分析結合Swiss Prot蛋白數(shù)據(jù)庫檢索,共鑒定了24個差異表達的蛋白質與生理鹽水注射(對照)比較,側腦室微量注射瘦素引起22個蛋白質分子顯著上調,2個顯著下調.重要的是,在上調的蛋白質分子中包括轉鐵蛋白、基質金屬蛋白酶7、膜聯(lián)蛋白A1、補體B因子、磷酸甘油酸酯變位酶1、烯醇酶、冠蛋白-1A等15個蛋白質分子,下調的2個蛋白質分子是補體C3、溶解物載體家族4成員3.這些蛋白質分子涉及細胞分化、細胞遷移、細胞凋亡,以及炎癥反應等功能.實驗結果提示,瘦素可能通過下丘腦-垂體軸參與免疫調節(jié)、雌性生殖及炎癥反應等,從而對子宮發(fā)揮調節(jié)作用.本研究將為加深對瘦素調節(jié)子宮作用的認識,并為深入研究女性生殖系統(tǒng)疾病提供新的啟示.
[Abstract]:Leptin (leptin) regulates reproductive activity through the hypothalamus-pituitary axis in animals, but the role of leptin in regulating uterus through hypothalamus is still unclear. Leptin was isolated by two-dimensional gel electrophoresis (2-DE) combined with matrix-assisted laser desorption / ionization time-of-flight mass spectrometry (MALDI-TOF-MS) 6 hours after microinjection of leptin into the lateral ventricle of rats. The protein expression profile of uterine fluid after ovariectomy with estrogen was identified. We obtained the 2-DE map with high resolution and good repeatability. Compared with saline injection (control), 24 differentially expressed proteins were identified by mass spectrometry combined with Swiss Prot protein database search. Twenty-two protein molecules were up-regulated and 2 significantly down-regulated by microinjection of leptin into lateral ventricle. Importantly, the up-regulated protein molecules include transferrin, matrix metalloproteinases 7, membranous protein A1, complement B factor, phosphoglycerate transposase 1, enolase, crown-1A, and so on. The down-regulated two protein molecules are complement C3, 4 members of the soluble carrier family 3. These proteins are involved in cell differentiation, cell migration, apoptosis, and inflammatory responses. The results suggest that leptin may be involved in immunomodulation, female reproduction and inflammatory response through hypothalamus-pituitary axis, which may play a regulatory role in uterus. This study will deepen the understanding of the role of leptin in the regulation of uterus, and provide a new inspiration for the further study of female reproductive system diseases.
