【摘要】：目的 研究轉(zhuǎn)錄因子激活蛋白-2α(Transcription factor activator protein-2α,AP-2α)對(duì)滋養(yǎng)細(xì)胞Be Wo凋亡、侵襲等生物學(xué)功能的影響,探討在子癇前期(preeclampsia,PE)發(fā)生發(fā)展中的作用。方法 將已構(gòu)建的p IRES2-EGFP/AP-2α真核表達(dá)載體,采用瞬時(shí)轉(zhuǎn)染的方法轉(zhuǎn)入滋養(yǎng)細(xì)胞系Be Wo細(xì)胞中,應(yīng)用real timePCR及Western blot方法,檢測(cè)各組Be Wo細(xì)胞中AP-2αm RNA及蛋白表達(dá);采用流式細(xì)胞儀檢測(cè)細(xì)胞凋亡,Transwell法檢測(cè)轉(zhuǎn)染后各組細(xì)胞侵襲力。結(jié)果 AP-2α轉(zhuǎn)染Be Wo細(xì)胞后AP-2αm RNA和蛋白表達(dá)量較對(duì)照組顯著增加,其差異有統(tǒng)計(jì)學(xué)意義(P0.05)。流式細(xì)胞儀檢測(cè)細(xì)胞凋亡結(jié)果顯示:與對(duì)照組比較,AP-2α轉(zhuǎn)染組細(xì)胞凋亡數(shù)量明顯增多,差異有統(tǒng)計(jì)學(xué)意義(P0.05);Transwell檢測(cè)細(xì)胞侵襲能力實(shí)驗(yàn)結(jié)果顯示:與對(duì)照組比較,AP-2α轉(zhuǎn)染組細(xì)胞侵襲的數(shù)量顯著減少,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論 證明AP-2α促進(jìn)滋養(yǎng)細(xì)胞凋亡并降低其侵襲力,造成滋養(yǎng)細(xì)胞層淺表植入,從而為子癇前期致病機(jī)理研究提供了理論依據(jù)。
[Abstract]:Objective to investigate the effects of transcription factor activator protein-2 偽 (Transcription factor activator protein-2 偽 (AP-2 偽) on the apoptosis and invasion of trophoblastic Be Wo and its role in the development of preeclampsia (preeclampsia,PE). Methods the constructed eukaryotic expression vector of p IRES2-EGFP/AP-2 偽 was transfected into trophoblastic Be Wo cells by transient transfection. The expression of AP-2 偽 m RNA and protein in Be Wo cells was detected by real timePCR and Western blot methods. Apoptosis was detected by flow cytometry and invasiveness was detected by Transwell assay. Results the expression of AP-2 偽 m RNA and protein in Be Wo cells transfected with AP-2 偽 was significantly higher than that in control group (P0.05). The results of flow cytometry showed that the number of apoptosis in AP-2 偽 transfected group was significantly higher than that in control group (P0.05). Compared with the control group, the invasiveness of AP-2 偽 transfected group was significantly decreased by Transwell (P0.05), and the invasiveness of the transfected group was significantly lower than that of the control group (P0.05). Conclusion AP-2 偽 promotes trophoblast apoptosis and reduces the invasion of trophoblast, which results in superficial implantation of trophoblast layer, which provides a theoretical basis for the study of pathogenesis of preeclampsia.
