【摘要】：目的：復(fù)發(fā)性自然流產(chǎn)是妊娠初期最常見(jiàn)的并發(fā)癥之一，其病因及發(fā)病機(jī)制及極繁雜，目前臨床上尚無(wú)明確有效的預(yù)測(cè)和防治措施。本研究旨在探討血管內(nèi)皮生長(zhǎng)因子(VEGF)的基因位點(diǎn)㧟2578C/A、㧟1154G/A、㧟634G/C、㧟460C/T和㧏936C/T的單核甘酸多態(tài)性(SNPs)與廣西地區(qū)壯族婦女復(fù)發(fā)性自然流產(chǎn)(RSA)患者發(fā)病的關(guān)系，從基因水平探討RSA發(fā)病的基因易感性，尋求與RSA相關(guān)的VEGF易感基因，進(jìn)一步闡明RSA發(fā)病的分子生物學(xué)機(jī)制，為研究RSA的發(fā)病機(jī)理提供一種可行性的方法。 方法：病例組選取門(mén)診就診的復(fù)發(fā)性流產(chǎn)患者共120例，對(duì)照組選取門(mén)診就診的健康經(jīng)產(chǎn)妊娠婦女和住院待產(chǎn)的健康經(jīng)產(chǎn)妊娠婦女共110例，兩組入選觀察對(duì)象均抽取靜脈血，并采用傳統(tǒng)手工苯酚-氯仿法提取全血DNA，采用PCR-限制性片段長(zhǎng)度多態(tài)性方法(PCR-RFLP)檢測(cè)兩組觀察對(duì)象VEGF基因5個(gè)多態(tài)位點(diǎn)的SNPs基因型的表達(dá)和頻率分布情況。 結(jié)果： VEGF㧟2578C/A、㧟1154G/A、㧟634G/C、㧟460C/T和㧏936C/T5個(gè)位點(diǎn)SNPs的各個(gè)基因型和等位基因頻率表達(dá)分布在RSA組和對(duì)照組間差異均不顯著，沒(méi)有統(tǒng)計(jì)學(xué)意義(P＞0.05)。 結(jié)論： VEGF㧟2578C/A、㧟1154G/A、㧟634G/C、㧟460C/T和㧏936C/T5個(gè)位點(diǎn)的基因多態(tài)性可能與廣西地區(qū)壯族婦女復(fù)發(fā)性自然流產(chǎn)的發(fā)病易感性不相關(guān)聯(lián)。研究結(jié)果與部分研究不同可能是多態(tài)性位點(diǎn)在不同地區(qū)，不同種族、民族，不同人群中的作用不一致所致。
[Abstract]:Objective: recurrent spontaneous abortion is one of the most common complications in early pregnancy. The aim of this study was to investigate the gene locus of vascular endothelial growth factor (VEGF), 2578 C / A, 1154 G / A, 634 G / C, ? 460C/T and? 936C/T mononuclear glycosylated (SNPs) were associated with the incidence of recurrent spontaneous abortion (RSA) in Zhuang women in Guangxi. The genetic susceptibility of RSA was investigated at the gene level, and the VEGF susceptibility genes associated with RSA were sought. Further elucidation of molecular biological mechanism of RSA provides a feasible method for studying the pathogenesis of RSA. Methods: 120 patients with recurrent abortion were selected in the case group, 110 healthy pregnant women in the outpatient department and 110 healthy pregnant women in the hospital were selected in the control group. Venous blood was drawn from the two groups. Whole blood DNA, was extracted by traditional manual phenol-chloroform method and PCR- restriction fragment length polymorphism (PCR-RFLP) was used to detect the expression and frequency distribution of SNPs genotypes in 5 polymorphic loci of VEGF gene in two groups. Results: the frequencies of SNPs genotypes and alleles in VEGF?2578C/A,?1154G/A,?634G/C,?460C/T and? 936C/T5 loci were not significantly different between RSA group and control group. There was no statistical significance (P > 0.05). Conclusion: the genetic polymorphisms of VEGF?2578C/A,?1154G/A,?634G/C,?460C/T and? 936C/T5 loci may not be associated with the susceptibility to recurrent spontaneous abortion in Zhuang women in Guangxi. The difference between the results and some studies may be due to the different roles of polymorphic loci in different regions, races, ethnic groups and different populations.
