【摘要】：目的:研究線粒體解耦聯(lián)蛋白2(UCP2)對(duì)宮頸鱗癌增殖、凋亡、轉(zhuǎn)移的影響。方法:通過(guò)質(zhì)粒轉(zhuǎn)染技術(shù),下調(diào)宮頸癌細(xì)胞系中UCP2的表達(dá),分別用MTT、流式細(xì)胞術(shù)、Transwell小室檢測(cè)宮頸鱗癌細(xì)胞增殖、凋亡、侵襲、轉(zhuǎn)移能力的變化;利用生物信息學(xué)的方法對(duì)UCP2蛋白進(jìn)行生物信息學(xué)分析,并分析與之相互作用的蛋白。結(jié)果:(1)q RT-PCR和WB證實(shí)質(zhì)粒轉(zhuǎn)染后UCP2在宮頸癌細(xì)胞株中的表達(dá)較對(duì)照組明顯降低。(2)MTT、凋亡、Transwell小室實(shí)驗(yàn)結(jié)果顯示,下調(diào)UCP2后Si Ha細(xì)胞的增殖能力明顯減弱,凋亡率增加,轉(zhuǎn)移能力減弱(P0.05)。(3)生物信息學(xué)分析顯示UCP2蛋白定位于線粒體,可能存在多個(gè)磷酸化位點(diǎn),且與多種蛋白相互作用,發(fā)揮生物學(xué)效應(yīng)。結(jié)論:UCP2可能在宮頸癌的發(fā)生和發(fā)展中發(fā)揮重要作用,有望成為宮頸癌的早期診斷標(biāo)志物和治療的新靶點(diǎn)。
[Abstract]:Aim: to study the effects of mitochondrial decoupling protein 2 (UCP2) on proliferation, apoptosis and metastasis of cervical squamous cell carcinoma. Methods: the expression of UCP2 in cervical carcinoma cell line was down-regulated by plasmid transfection technique. The proliferation, apoptosis, invasion and metastasis of cervical squamous cell carcinoma cells were detected by MTT, flow cytometry and Transwell chamber. UCP2 protein was analyzed by bioinformatics and the proteins interacting with it were analyzed. Results: (1) Q RT-PCR and WB confirmed that the expression of UCP2 in cervical cancer cell lines after transfection was significantly lower than that in the control group. (2) MTT, apoptosis. The results of Transwell chamber experiment showed that the proliferation ability of Si Ha cells decreased significantly after down-regulation of UCP2. The bioinformatics analysis showed that the UCP2 protein was located in mitochondria with multiple phosphorylation sites and interacted with many proteins to play a biological effect. Conclusion: UCP2 may play an important role in the occurrence and development of cervical cancer and may be a new target for early diagnosis and treatment of cervical cancer.
