宮頸鱗癌組織中HPV16 E6與人端粒保護(hù)蛋白1的檢測及關(guān)系的研究

【摘要】:目的:研究宮頸鱗癌、宮頸上皮內(nèi)瘤樣病變(Cervical intraepithelial neoplasia,CIN)及正常宮頸組織中人乳頭瘤病毒(Human papilloma virus,HPV)16 E6蛋白及人端粒保護(hù)蛋白1(Human protection of telomeres 1,POT1)的表達(dá)情況及關(guān)系。方法:收集山西醫(yī)科大學(xué)附屬第一醫(yī)院2013年5月至2016年5月期間10例正常宮頸組織(無HPV感染)、30例宮頸上皮內(nèi)瘤樣病變組織(其中CINI5例、CINII10例、CINIII15例)、40例宮頸鱗癌組織。采用免疫組織化學(xué)SP方法分別檢測以上宮頸組織中HPV16 E6蛋白和POT1蛋白的表達(dá)情況及其相關(guān)性。結(jié)果:1.POT1蛋白在正常宮頸組、CIN組、宮頸鱗癌組織的表達(dá)呈現(xiàn)出逐漸下降的趨勢,三組間兩兩比較差異有統(tǒng)計學(xué)意義(χ2=19.662,P0.05)。HPV16 E6蛋白在正常宮頸組、CIN組、宮頸鱗癌組織的表達(dá)呈現(xiàn)出逐漸上升的趨勢,差異有統(tǒng)計學(xué)意義(χ2=26.362,P0.05)。2.在不同年齡、臨床分期、腫瘤大小、組織分化程度的宮頸鱗癌組織中HPV16 E6蛋白和POT1蛋白表達(dá)差異無統(tǒng)計學(xué)意義(P0.05),宮頸鱗癌組織中POT1在無淋巴轉(zhuǎn)移的表達(dá)高于有淋巴轉(zhuǎn)移者,差異有統(tǒng)計學(xué)意義(χ2=7.677,P=0.0270.05),HPV16E6蛋白的表達(dá)與淋巴轉(zhuǎn)移有關(guān),差異有統(tǒng)計學(xué)意義(χ2=4.912,P0.05)。3.運用Spearman相關(guān)性分析顯示HPV16 E6蛋白和POT1蛋白陽性表達(dá)呈負(fù)相關(guān)(rs=-0.565,P0.01)。結(jié)論:宮頸鱗癌組織中HPV16 E6蛋白與POT1蛋白陽性表達(dá)呈負(fù)相關(guān),其機制可能是通過HPV16 E6蛋白的表達(dá)抑制了POT1蛋白的表達(dá),進(jìn)而促進(jìn)宮頸鱗癌的發(fā)生、發(fā)展。
[Abstract]:Objective: to study the expression of human papillomavirus (Human papilloma virus,HPV) 16 E6 protein and human telomere protection protein 1 (Human protection of telomeres 1 in cervical squamous cell carcinoma, cervical intraepithelial neoplasia (Cervical intraepithelial neoplasia,CIN) and normal cervical tissues. The expression and relationship of POT1. Methods: from May 2013 to May 2016, 10 cases of normal cervical tissues (without HPV infection) and 30 cases of cervical intraepithelial neoplasia (CINI5, CINII10, CINIII15) were collected from the first affiliated Hospital of Shanxi Medical University. 40 cases of cervical squamous cell carcinoma. Immunohistochemical SP method was used to detect the expression of HPV16 E6 protein and POT1 protein in cervical tissues. Results: the expression of 1.POT1 protein in normal cervix group, CIN group and cervical squamous cell carcinoma showed a decreasing trend, and there was a significant difference among the three groups (蠂 2 + 19.662P 0.05). HPV16 E6 protein was expressed in normal cervix group and CIN group. The expression of squamous cell carcinoma of the cervix showed an increasing trend, and the difference was statistically significant (蠂 ~ 2 ~ 2 ~ (26.362) P _ (0.05). There was no significant difference in the expression of HPV16 E6 protein and POT1 protein in cervical squamous cell carcinoma tissues with different age, clinical stage, tumor size and tissue differentiation (P0.05). The expression of POT1 in cervical squamous cell carcinoma was significantly higher than that in patients without lymphatic metastasis (蠂 27.677Pu 0.0270.05). The expression of HPV16E6 protein was related to lymphatic metastasis (蠂 2 + 4.912). P0.05) The positive expression of HPV16 E6 protein and POT1 protein was negatively correlated (rs=-0.565,P0.01) by Spearman correlation analysis. Conclusion: there is a negative correlation between HPV16 E6 protein and POT1 protein expression in squamous cell carcinoma of cervix. The mechanism may be that HPV16 E6 protein inhibits the expression of POT1 protein and promotes the occurrence and development of cervical squamous cell carcinoma.
