【摘要】：目的： 通過觀察白藜蘆醇(Res)對(duì)人卵巢癌Skov3細(xì)胞的增殖抑制作用及其對(duì)Sirt1表達(dá)的影響，初步探討Res對(duì)人卵巢癌Skov3細(xì)胞的生長(zhǎng)抑制作用的相關(guān)機(jī)制，為卵巢癌的治療提供實(shí)驗(yàn)和理論依據(jù)。 方法： 體外培養(yǎng)人卵巢癌Skov3細(xì)胞，用不同濃度（分別是0、5、10、20、40、80μg/ml）的Res作用于Skov3細(xì)胞24h、48h、72h，應(yīng)用MTT方法檢測(cè)不同濃度的藥物對(duì)人卵巢癌Skov3細(xì)胞增殖的影響；應(yīng)用RT-PCR和Western blot方法檢測(cè)不同濃度的Res作用于人卵巢癌Skov3細(xì)胞24h后其Sirt1mRNA和蛋白表達(dá)水平的變化。 結(jié)果： 1.不同濃度的Res（0、5、10、20、40、80μg/ml）作用于人卵巢癌Skov3細(xì)胞不同時(shí)間（24h、48h、72h）后，，人卵巢癌Skov3細(xì)胞的生長(zhǎng)受到抑制，且抑制作用隨時(shí)間和濃度的增加而增強(qiáng)，組與組之間的比較及組與對(duì)照組間的比較，差異均有統(tǒng)計(jì)學(xué)意義（P0.05）； 2.不同濃度的Res（0、5、10、20、40、80μg/ml）作用于人卵巢癌Skov3細(xì)胞24h，隨著藥物濃度的增加，人卵巢癌Skov3細(xì)胞中Sirt1mRNA的表達(dá)逐漸下調(diào)（P0.05）； 3.不同濃度的Res（0、5、10、20、40、80μg/ml）作用于人卵巢癌Skov3細(xì)胞24h，隨著藥物濃度的增加，人卵巢癌Skov3的Sirt1蛋白的表達(dá)逐漸下調(diào)（P0.05）。 結(jié)論： 1. Res能抑制人卵巢癌Skov3細(xì)胞的增殖，且抑制作用存在時(shí)間濃度依賴關(guān)系。 2. Res可能是通過下調(diào)Sirt1的表達(dá)發(fā)揮對(duì)人卵巢癌Skov3細(xì)胞的抑制作用。
[Abstract]:Aim: to investigate the inhibitory effect of resveratrol (Res) on the proliferation of human ovarian cancer Skov3 cells and the expression of Sirt1, and to explore the mechanism of Res on the growth inhibition of human ovarian cancer Skov3 cells. To provide experimental and theoretical basis for the treatment of ovarian cancer. Methods: human ovarian cancer Skov3 cells were cultured in vitro and treated with different concentrations of Res (0 ~ (51010) ~ 20 ~ 4080 渭 g/ml) for 24 h ~ 48 h ~ 72 h. The effects of different concentrations of drugs on the proliferation of human ovarian cancer Skov3 cells were detected by MTT method. RT-PCR and Western blot methods were used to detect the expression of Sirt1mRNA and protein in human ovarian cancer Skov3 cells treated with different concentrations of Res for 24 hours. Results: 1. The growth of human ovarian cancer Skov3 cells was inhibited by different concentrations of Res (0H5, 1020, 4080 渭 g/ml) at different time (24 h, 48 h, 72 h), and the inhibitory effect increased with the increase of time and concentration. The differences between the two groups and between the two groups were statistically significant (P0.05). 2. The expression of Sirt1mRNA in human ovarian cancer Skov3 cells decreased gradually with the increase of drug concentration (P 0.05). The expression of Sirt1 protein in human ovarian cancer Skov3 cells decreased gradually with the increase of drug concentration (P05). Conclusion: 1. Res can inhibit the proliferation of human ovarian cancer Skov3 cells in a time and concentration dependent manner. 2. Res may play an inhibitory role on human ovarian cancer Skov3 cells by down-regulating the expression of Sirt1.
