甲狀腺激素受體基因與卵巢癌順鉑耐藥關(guān)系的研究

【摘要】:目的探討甲狀腺激素受體基因(THRB)是否為耐藥相關(guān)基因或影響卵巢癌細(xì)胞耐藥。方法合成特異性基因探針,利用實時熒光定量聚合酶鏈反應(yīng)(qRT-PCR)技術(shù)顯示細(xì)胞在激素作用下的基因表達(dá)差異。結(jié)果 THRB的表達(dá)在人卵巢癌耐順鉑SKOV3/DDP細(xì)胞中的表達(dá)量高于非耐藥卵巢癌SKOV3細(xì)胞。結(jié)論甲狀腺激素能促進(jìn)人卵巢癌SKOV3細(xì)胞和人卵巢癌SKOV3/DDP耐順鉑細(xì)胞的增殖,THRB在卵巢癌耐藥SKOV3/DDP細(xì)胞中的表達(dá)的量遠(yuǎn)高于一般卵巢癌SKOV3細(xì)胞,THRB對卵巢癌耐藥起著非常重要的作用,可能是耐藥相關(guān)基因,其機制有必要進(jìn)一步探討。
[Abstract]:Objective to investigate whether thyroid hormone receptor gene (THRB) is a drug resistance related gene or affects ovarian cancer cell resistance. Methods the specific gene probe was synthesized and the difference of gene expression in the cells under the action of hormone was demonstrated by real-time fluorescence quantitative polymerase chain reaction (qRT-PCR). Results the expression of THRB in cisplatin-resistant SKOV3/DDP cells was higher than that in non-drug-resistant ovarian cancer SKOV3 cells. Conclusion thyroid hormone can promote the proliferation of human ovarian cancer SKOV3 cells and human ovarian cancer SKOV3/DDP cisplatin resistant cells. The expression of THRB in drug-resistant SKOV3/DDP cells of ovarian cancer is much higher than that of normal ovarian cancer SKOV3 cells. THRB plays an important role in drug resistance of ovarian cancer. It may be a drug resistance related gene, and its mechanism needs to be further explored.
