【摘要】：目的探討卵丘顆粒細(xì)胞高爾基磷酸化蛋白-3(GOLPH3)表達(dá)對單精子卵胞漿內(nèi)注射(ICSI)助孕結(jié)局的影響。方法收集2012年4月~2014年6月,因男性不育癥行ICSI助孕的119例女性患者的卵丘顆粒細(xì)胞,采用細(xì)胞免疫化學(xué),Western bloting方法檢測顆粒細(xì)胞GOLPH3蛋白的表達(dá),采用real-time熒光定量PCR方法檢測GOLPH3基因表達(dá)。比較分析ICSI妊娠組與非妊娠組病例卵丘顆粒細(xì)胞GOLPH3表達(dá)率差異,并分析它們與ICSI實(shí)驗(yàn)室結(jié)局各項(xiàng)指標(biāo):穿卵泡數(shù)、獲卵數(shù)、獲卵率;受精數(shù)、受精率;卵裂數(shù)、卵裂率;優(yōu)質(zhì)胚胎數(shù)、囊胚形成數(shù)及囊胚冷凍數(shù)之間的相關(guān)關(guān)系。結(jié)果對119份卵丘顆粒細(xì)胞涂片進(jìn)行免疫細(xì)胞化學(xué)分析GOLPH3的表達(dá),結(jié)果發(fā)現(xiàn)GOLPH3主要表達(dá)在細(xì)胞質(zhì)中,妊娠組與未妊娠組卵丘顆粒細(xì)胞GOLPH3表達(dá)率與表達(dá)強(qiáng)度組間差異均有統(tǒng)計學(xué)意義(P均0.05)。卵丘顆粒細(xì)胞GOLPH3表達(dá)率與ICSI實(shí)驗(yàn)室結(jié)局指標(biāo):穿卵泡數(shù),Ⅲ級OCCs數(shù),ICSI卵子數(shù),受精數(shù),卵裂數(shù),平均優(yōu)質(zhì)胚胎數(shù),平均囊胚數(shù),優(yōu)質(zhì)囊胚數(shù),平均冷凍胚胎數(shù)等呈顯著正相關(guān)關(guān)系(P均0.01)。Real-Time PCR,Western blot檢測妊娠組與未妊娠組顆粒細(xì)胞GOLPH3蛋白與基因表達(dá)組間差異也均有統(tǒng)計學(xué)意義(P均0.05)。結(jié)論 GOLPH3在卵丘顆粒細(xì)胞的表達(dá),可能影響卵母細(xì)胞的成熟,卵子的發(fā)育能力,進(jìn)而與ICSI助孕結(jié)局有關(guān)。
[Abstract]:Objective to investigate the effect of Golgi phosphorylated protein-3 (GOLPH3) expression in cumulus granulosa cells on the outcome of pregnancy assisted by intracytoplasmic injection of (ICSI). Methods from April 2012 to June 2014, the cumulus granulosa cells were collected from 119 female patients with ICSI assisted pregnancy due to male infertility. The expression of GOLPH3 protein in granulosa cells was detected by cytoimmunochemical, Western bloting assay. Real-time fluorescence quantitative PCR was used to detect the expression of GOLPH3 gene. To compare and analyze the difference of GOLPH3 expression rate of cumulus granulosa cells between ICSI pregnancy group and non-pregnancy group, and to analyze the indexes of ICSI laboratory outcome: the number of follicles pierced, the rate of obtaining eggs, the number of fertilization, the rate of fertilization, the number of cleavage, the rate of cleavage, the percentage of cleavage, the rate of cleavage. The correlation among the number of high quality embryos, the number of blastocysts and the number of frozen blastocysts. Results the expression of GOLPH3 was detected by immunocytochemistry in 119 smears of cumulus granulosa cells. The results showed that GOLPH3 was mainly expressed in cytoplasm.
