缺氧下5-脂氧合酶在卵巢癌中的表達(dá)及意義

【摘要】:目的:檢測(cè)缺氧環(huán)境下5-LOX在卵巢癌中的表達(dá)及與臨床參數(shù)間的關(guān)系,探討其在卵巢癌發(fā)生發(fā)展及治療中的意義。方法:免疫組化檢測(cè)56例卵巢癌組織中5-LOX與HIF-1α表達(dá),并分析5-LOX表達(dá)與卵巢癌臨床參數(shù)之間的關(guān)系。體外將人卵巢癌SKOV3細(xì)胞在缺氧環(huán)境下培養(yǎng)0、12、24h和48h,分別提取RNA和蛋白。qRT-PCR法和Western blot法分別檢測(cè)5-LOX mRNA和蛋白表達(dá);ELISA法檢測(cè)缺氧環(huán)境下卵巢癌細(xì)胞5-LOX代謝產(chǎn)物5-HETE的變化。結(jié)果:5-LOX與HIF-1α表達(dá)具有明顯的相關(guān)性,其表達(dá)水平與患者的淋巴結(jié)轉(zhuǎn)移、遠(yuǎn)處轉(zhuǎn)移和臨床分期密切相關(guān)。缺氧條件下培養(yǎng)的SKOV3細(xì)胞的5-LOX在轉(zhuǎn)錄和蛋白水平均表達(dá)升高;缺氧條件下卵巢癌細(xì)胞培養(yǎng)上清的5-HETE較常氧條件下明顯升高。結(jié)論:缺氧能促進(jìn)卵巢癌5-LOX表達(dá)及增加代謝產(chǎn)物5-HETE的產(chǎn)生,提示5-LOX可作為治療卵巢癌的潛在靶點(diǎn),這為卵巢癌的發(fā)病機(jī)理研究和診治提供了新的思路與方法。
[Abstract]:Objective: to detect the expression of 5-LOX in ovarian cancer under hypoxia and its relationship with clinical parameters, and to explore its significance in the development and treatment of ovarian cancer. Methods: the expression of 5-LOX and HIF-1 偽 was detected by immunohistochemistry in 56 cases of ovarian cancer, and the relationship between 5-LOX expression and clinical parameters of ovarian cancer was analyzed. Human ovarian cancer SKOV3 cells were cultured in hypoxic environment for 24 h and 48 h respectively to extract RNA and protein. QRT-PCR and Western blot methods were used to detect the expression of 5-LOX mRNA and protein respectively. ELISA method was used to detect the changes of 5-LOX metabolites 5-HETE in hypoxic environment. Results: the expression of 5-LOX and HIF-1 偽 was significantly correlated with lymph node metastasis, distant metastasis and clinical stage. The expression of 5-LOX in cultured SKOV3 cells under hypoxia was higher than that in normal oxygen, while the expression of 5-HETE in the supernatant of ovarian cancer cells cultured under hypoxia was significantly higher than that in normal oxygen. Conclusion: hypoxia can promote the expression of 5-LOX and increase the production of metabolite 5-HETE in ovarian cancer, suggesting that 5-LOX can be used as a potential target for the treatment of ovarian cancer, which provides a new idea and method for the study of pathogenesis and diagnosis and treatment of ovarian cancer.
