miR-100在上皮性卵巢癌順鉑耐藥細(xì)胞中的表達(dá)及其與耐藥的關(guān)系

【摘要】:目的研究微小RNA-100(mi R-100)在人上皮性卵巢癌順鉑耐藥細(xì)胞中的表達(dá)及其與順鉑耐藥的關(guān)系。方法脂質(zhì)體lipofectamine 2000介導(dǎo)成熟mi R-100模擬物(mimics)及陰性對(duì)照片段(NC)、阻遏物(inhibitor)及陰性對(duì)照片段(inhibitor NC)分別轉(zhuǎn)染人卵巢癌順鉑耐藥細(xì)胞株SKOV3/DDP。實(shí)驗(yàn)分為6組:SKOV3組、SKOV3/DDP組、mi R-100 mimics組、NC組、mi R-100 inhibitor組及inhibitor NC組。實(shí)時(shí)熒光定量PCR及CCK8法分別檢測各組細(xì)胞中mi R-100的表達(dá)和順鉑半數(shù)抑制濃度(IC50)。結(jié)果(1)SKOV3/DDP細(xì)胞的順鉑耐藥指數(shù)(RI)為2.23;(2)mi R-100在SKOV3/DDP細(xì)胞中低表達(dá),與SKOV3相比差異有統(tǒng)計(jì)學(xué)意義(P0.001);(3)轉(zhuǎn)染mi R-100 mimics后,SKOV3/DDP細(xì)胞中mi R-100的表達(dá)水平與NC組相比上升38.29倍,差異有統(tǒng)計(jì)學(xué)意義(P0.01);mi R-100 mimics組順鉑IC50與NC組相比明顯降低,差異有統(tǒng)計(jì)學(xué)意義(P0.001);(4)轉(zhuǎn)染mi R-100 inhibitor后,SKOV3/DDP細(xì)胞中mi R-100的表達(dá)水平與inhibitor NC組相比下降97.7%(P0.001);mi R-100 inhibitor組順鉑IC50與inhibitor NC組相比明顯升高(P0.001)。結(jié)論 mi R-100在人卵巢癌順鉑耐藥細(xì)胞中低表達(dá),mi R-100可增加卵巢癌順鉑耐藥細(xì)胞株對(duì)順鉑的敏感性,從而逆轉(zhuǎn)其耐藥。
[Abstract]:Objective to investigate the expression of small RNA-100 (mi R-100) in cisplatin resistant human epithelial ovarian cancer cells and its relationship with cisplatin resistance. Methods Liposome lipofectamine 2000 mediated transfection of mature mi R-100 mimetic (mimics), negative control (NC), repressor (inhibitor) and negative control (inhibitor NC) into human ovarian cancer cell line SKOV3/DDP. The experiment was divided into 6 groups: SKOV3 group, SKOV3 / DDP group, Mi R-100 mimics group, NC group, Mi R-100 inhibitor group and inhibitor NC group. The expression of mi R-100 and the half inhibitory concentration (IC50) of cisplatin were detected by real-time fluorescence quantitative PCR and CCK8 respectively. Results (1) the cisplatin resistance index (RI) of SKOV3/DDP cells was 2.23; (2) the expression of mi R-100 was lower in SKOV3/DDP cells than that in SKOV3 (P0.001); (3). The expression level of mi R-100 in SKOV3 / DDP cells was 38.29 times higher than that in NC group after mi R-100 mimics transfection. The difference was statistically significant (P0.01). The expression of mi R-100 in mi R-100 mimics group was significantly lower than that in NC group (P0.001); (4). After transfection of mi R-100 inhibitor, the expression of mi R-100 in SKOV3 / DDP cells was significantly decreased by 97.7% (P0.001) compared with that in inhibitor NC group (P0.001). The expression of cisplatin IC50 in inhibitor group was significantly higher than that in inhibitor NC group (P0.001). Conclusion the low expression of mi R-100 in cisplatin resistant human ovarian cancer cells can increase the sensitivity of cisplatin resistant ovarian cancer cell line to cisplatin and reverse its drug resistance.
