神經(jīng)節(jié)苷脂GM3對人宮頸癌HeLa細胞凋亡和血管內(nèi)皮生長因子表達的影響

【摘要】:目的研究外源性神經(jīng)節(jié)苷脂GM3對人宮頸癌He La細胞增殖、凋亡和VEGF表達的影響。方法不同濃度GM3干預HeLa細胞24 h后,MTT法檢測細胞增殖變化,流式細胞學技術(shù)檢測細胞凋亡率,RT-PCR技術(shù)檢測細胞VEGF mRNA表達水平,Western blot技術(shù)檢測細胞VEGF蛋白水平。結(jié)果(1)GM3濃度分別為2、10、50和250μmol/L時,HeLa細胞生長抑制率分別為9.8%、32.9%、50.7%和78.6%,半數(shù)抑制濃度(IC50)為49.8μmol/L。(2)GM3濃度分別為10、50和250μmol/L時,HeLa細胞凋亡率分別為(4.9±0.4)%、(15.1±0.3)%、(24.4±0.7)%。(3)當GM3濃度高于10μmol/L時,HeLa細胞VEGF mRNA表達水平以及VEGF蛋白表達水平均顯著下降(P0.05)。結(jié)論外源性神經(jīng)節(jié)苷脂GM3能呈濃度依賴性抑制人宮頸癌He La細胞增殖,促進HeLa細胞凋亡,下調(diào)HeLa VEGF mRNA水平和蛋白水平,提示GM3可能通過調(diào)節(jié)腫瘤細胞凋亡和腫瘤血管生成而發(fā)揮雙重抗腫瘤作用。
[Abstract]:Objective to study the effects of exogenous ganglioside GM3 on proliferation, apoptosis and VEGF expression of human cervical cancer He La cells. Methods the proliferation of HeLa cells was detected by MTT assay after 24 h intervention with different concentrations of GM3. Apoptosis rate was detected by flow cytometry and the expression level of VEGF mRNA was detected by RT-PCR. VEGF protein was detected by Western blot. Results (1) the growth inhibition rates of HeLa cells at GM3 concentration of 210U 50 and 250 渭 mol/L were 9.8and 32.9% and 78.6%, respectively. The half inhibitory concentration (IC50) was 49.8 渭 mol/L. (2) and the GM3 concentration was 109.50 渭 mol/L. (2) and 250 渭 mol/L, respectively. The apoptotic rate was (4.9 鹵0.4), (15.1 鹵0.3), (24.4 鹵0.7) when GM3 concentration was higher than 10 渭 mol/L. (3) when GM3 concentration was higher than 10 渭 mol/L, the apoptotic rate of Hela cells was (4.9 鹵0.4), (24.4 鹵0.7), respectively. (3) when the GM3 concentration was higher than 10 渭 mol/L, the apoptotic rate was (4.9 鹵0.4), (24.4 鹵0.7), respectively. The level of VEGF protein expression decreased significantly (P0.05). Conclusion exogenous ganglioside GM3 can inhibit the proliferation of human cervical cancer He La cells in a concentration-dependent manner, promote the apoptosis of HeLa cells, and down-regulate the level of HeLa VEGF mRNA and protein. The results suggest that GM3 may play a dual anti-tumor effect by regulating tumor cell apoptosis and tumor angiogenesis.
