戊酸雌二醇和黑升麻異丙醇提取物對卵巢切除小鼠的影響及其機(jī)制探討
[Abstract]:Objective to investigate the cardiovascular protection mechanism of estrogen, to evaluate the effects of isopropanol extract of black cohosh on ovariectomized mice, and to evaluate its safety and efficacy. Methods Twenty C57BL/6 mice and 20 APOE-/- mice were selected and bilateral ovariectomies were used to establish menopausal animal model. Each strain of mice was divided into four groups: sham-operated group, (SHAM NS), simple ovariectomized group, (OVX NS), ovariectomized with estrogen supplement group (OVX E2) and ovariectomized group supplemented with isopropanol extract group (OVX ICR). Body weight, blood lipid profile, aorta, reproductive system and nervous system were assessed after 12 weeks of intervention. Result 1. Weight and fat distribution) OVX NS,OVX ICR group was larger than SHAM NS,OVX E2 group (P0.05) OVX NS,OVX ICR group had more abdominal fat than SHAM NS,OVX E2 group (3) cholesterol level in OVX NS,OVX ICR group was higher than SHAM NS,OVX E2 group. The percentage of atherosclerotic plaque area in the NS,OVX ICR group was higher than that in the SHAM NS,OVX E2 group (P0.05). Uterine weight was smaller than that in SHAM NS,OVX E2 group (P0.05). The immobility time of OVX NS group was longer than that of SHAM NS,OVX E2OVX ICR group (P0.05). Conclusion 1. Estradiol supplementation can reduce the weight of ovariectomized mice, reduce abdominal fat, reduce the level of blood lipid, reduce the percentage of aortic atherosclerotic plaque area, and protect the cardiovascular system. The isopropanol extract of black cohosh has no effect on cardiovascular system protection. Estradiol could promote the proliferation of ovariectomized mouse reproductive system, but the extract of isopropanol did not. 3. Estradiol and Isopropanol extract could shorten the immobility time and improve the mental state of ovariectomized mice.
