人胎盤(pán)來(lái)源間充質(zhì)干細(xì)胞對(duì)妊娠期高血壓病細(xì)胞滋養(yǎng)細(xì)胞凋亡的影響:隨機(jī)對(duì)照實(shí)驗(yàn)方案

【摘要】:背景:細(xì)胞滋養(yǎng)細(xì)胞凋亡增加、浸潤(rùn)能力下降是妊娠期高血壓病發(fā)病的重要環(huán)節(jié)之一。人胎盤(pán)來(lái)源間充質(zhì)干細(xì)胞已經(jīng)在許多研究中顯示出了對(duì)損傷的修復(fù)作用,而且胎盤(pán)間充質(zhì)干細(xì)胞還具有一定的內(nèi)分泌功能,可通過(guò)自分泌或旁分泌細(xì)胞因子的方式作用于近、遠(yuǎn)處的組織。因此,嘗試移植人胎盤(pán)來(lái)源間充質(zhì)干細(xì)胞修復(fù)損傷的滋養(yǎng)細(xì)胞功能,進(jìn)而治療妊娠期高血壓病,有一定可行性。方法/設(shè)計(jì):隨機(jī)對(duì)照細(xì)胞學(xué)實(shí)驗(yàn)。收集人胎盤(pán)來(lái)源間充質(zhì)干細(xì)胞培養(yǎng)液,過(guò)濾后即為人胎盤(pán)來(lái)源間充質(zhì)干細(xì)胞條件培養(yǎng)基。培養(yǎng)人絨毛膜滋養(yǎng)層細(xì)胞系JEG-3細(xì)胞,隨機(jī)分為3組,對(duì)照組在培養(yǎng)液中加入體積分?jǐn)?shù)15%正常孕婦血清;妊娠期高血壓病模型組在培養(yǎng)液中加入體積分?jǐn)?shù)15%子癇前期重度患者的血清;人胎盤(pán)來(lái)源間充質(zhì)干細(xì)胞條件培養(yǎng)組在培養(yǎng)液中加入體積分?jǐn)?shù)15%子癇前期重度患者血清培養(yǎng)24 h后,更換為體積分?jǐn)?shù)15%子癇前期重度患者血清+人胎盤(pán)來(lái)源間充質(zhì)干細(xì)胞條件培養(yǎng)基。采用流式細(xì)胞儀檢測(cè)各組細(xì)胞的凋亡率。討論:實(shí)驗(yàn)從人胎盤(pán)來(lái)源間充質(zhì)干細(xì)胞對(duì)妊娠期高血壓病細(xì)胞滋養(yǎng)細(xì)胞功能的影響入手,可能為妊娠期高血壓疾病的治療找到一條細(xì)胞移植之路。倫理批準(zhǔn):研究方案取得沈陽(yáng)醫(yī)學(xué)院附屬中心醫(yī)院臨床研究倫理委員會(huì)的書(shū)面批準(zhǔn)。正常及子癇前期孕婦簽署知情同意書(shū)。
[Abstract]:Background: the increase of cytotrophoblast apoptosis and the decrease of infiltrating ability are one of the important links in the pathogenesis of pregnancy-induced hypertension. Human placenta derived mesenchymal stem cells have been shown to repair damage in many studies, and placental mesenchymal stem cells also have endocrine function, which can be acted on by autocrine or paracrine cytokines. Distant tissue Therefore, it is feasible to transplant human placenta derived mesenchymal stem cells to repair the damaged trophoblastic function and to treat hypertensive disorder complicating pregnancy. Methods / Design: a randomized controlled cytological experiment. Human placenta derived mesenchymal stem cell culture medium was collected and filtered, which was then conditioned medium for human placenta derived mesenchymal stem cells. Human chorionic trophoblastic cell line JEG-3 cells were cultured and randomly divided into three groups. The control group added 15% normal maternal serum into the culture medium. The serum of 15% volume fraction of severe preeclampsia was added to the culture medium in the gestational hypertension model group. In the human placenta derived mesenchymal stem cell conditioned culture group, 15% volume fraction of serum was added to the culture medium for 24 h. The medium was replaced with 15% volume fraction of mesenchymal stem cells derived from human placenta in patients with severe preeclampsia. The apoptotic rate of each group was measured by flow cytometry. Discussion: based on the effect of human placenta derived mesenchymal stem cells on the function of trophoblast cells in hypertensive disorder complicating pregnancy, we may find a way of cell transplantation for the treatment of hypertensive disorder complicating pregnancy. Ethical approval: the research program has been approved by the Clinical Research Ethics Committee of the affiliated Central Hospital of Shenyang Medical College. Normal and preeclampsia pregnant women sign informed consent.
