TCF7L2基因多態(tài)性與多囊卵巢綜合征易感性的meta分析

【摘要】:目的:探討TCF7L2rs7903146、rs290487基因多態(tài)性與多囊卵巢綜合征易感性的的關(guān)聯(lián)強(qiáng)度。 方法:制定檢索式,并通過(guò)計(jì)算機(jī)檢索PubMed、EMbase、Cochrane Library以及中國(guó)生物醫(yī)學(xué)文獻(xiàn)數(shù)據(jù)庫(kù)、中國(guó)知網(wǎng)、萬(wàn)方數(shù)據(jù)庫(kù)公開(kāi)發(fā)表的關(guān)于TCF7L2的基因多態(tài)性與多囊卵巢綜合征易感性的相關(guān)文獻(xiàn)。檢索時(shí)間自建庫(kù)起至2014年3月1日。對(duì)相關(guān)文獻(xiàn)進(jìn)行篩選、質(zhì)量評(píng)價(jià)、異質(zhì)性評(píng)價(jià)并提取相關(guān)病例對(duì)照研究基本資料,利用Revman5.2軟件進(jìn)行meta分析,計(jì)算合并的比值比(odds ratio,OR)及其95%可信區(qū)間(confidence interval,CI)分析兩者間的關(guān)聯(lián)強(qiáng)度。 結(jié)果:共納入文獻(xiàn)8篇,其中5篇涉及研究位點(diǎn)rs7903146,4篇研究rs290487位點(diǎn),前者共包括病例組1661例及對(duì)照組2866例,后者病例組1544例,對(duì)照組1134例。根據(jù)異質(zhì)性分析結(jié)果使用隨機(jī)效應(yīng)模型或固定效應(yīng)模型,合并數(shù)據(jù)結(jié)果示rs7903146基因多態(tài)性與多囊卵巢綜合征易感性的風(fēng)險(xiǎn)模型中均提示兩者間無(wú)相關(guān)性(OR:0.98.1.10:95%CI:0.85-0.91to1.13.1.41).而rs290487基因位點(diǎn)攜帶C等位基因者PCOS患病風(fēng)險(xiǎn)增加(OR:1.11-1.46:95%CI:0.94-1.14to1.31-1.89)。 結(jié)論:TCF7L2rs7903146基因多態(tài)性與多囊卵巢綜合征的遺傳易感性不存在相關(guān)性。TCF7L2rs290487基因多態(tài)性中攜帶C等位基因者多囊卵巢綜合征患病風(fēng)險(xiǎn)增高,對(duì)于疾病今后的預(yù)防、診斷及基因靶向治療提供了依據(jù)。
[Abstract]:Objective: To investigate the association between polymorphism of TCF7L2rs7903146, rs290487 gene and susceptibility to polycystic ovary syndrome (PCOS).
METHODS: PubMed, EMbase, Cochrane Library and Chinese Biomedical Literature Database, China HowNet and Wanfang Database were searched by computer. The relevant literatures about TCF7L2 gene polymorphism and susceptibility to polycystic ovary syndrome were published. The retrieval time was from the establishment of the database to March 1, 2014. Screening, quality evaluation, heterogeneity evaluation and extracting the basic data of case-control studies were performed. Revman 5.2 software was used for meta-analysis. The odds ratio (OR) and its 95% confidence interval (CI) were calculated to analyze the correlation strength between the two.
RESULTS: A total of 8 articles were included, 5 of which involved rs7903146 and 4 of which involved rs290487. The former included 1661 cases in the case group and 2866 cases in the control group. The latter included 1544 cases in the case group and 1134 cases in the control group. There was no correlation between sex and polycystic ovary syndrome (OR: 0.98.1.10:95% CI: 0.85-0.91 to 1.13.1.41). The risk of PCOS was increased in rs290487 locus carriers (OR: 1.11-1.46:95% CI: 0.94-1.14 to 1.31-1.89).
Conclusion: The polymorphism of TCF7L2rs7903146 gene is not associated with the genetic susceptibility to polycystic ovary syndrome.
