DAPT抑制妊娠滋養(yǎng)細(xì)胞腫瘤Notch信號(hào)通路對(duì)細(xì)胞增殖和EMT影響的研究

【摘要】:目的:探討Notch信號(hào)通路在人妊娠滋養(yǎng)細(xì)胞腫瘤發(fā)生發(fā)展過(guò)程中的重要作用,及其與EMT之間的關(guān)系。方法:采用γ-分泌酶抑制劑DAPT處理JAR和JEG-3兩種人絨毛膜癌細(xì)胞株,MTT法檢測(cè)兩種細(xì)胞株的增殖情況,q PCR法檢測(cè)Notch1 mRNA表達(dá)。Western blot法檢測(cè)DAPT阻斷Notch信號(hào)通路后JAR細(xì)胞中EMT相關(guān)蛋白E-cad的表達(dá)情況。結(jié)果:DAPT能抑制JEG-3、JAR細(xì)胞生長(zhǎng),10μmol/L和15μmol/L DAPT作用48h開(kāi)始分別抑制JEG-3和JAR細(xì)胞生長(zhǎng)(P0.05),且抑制作用呈時(shí)間和濃度依賴關(guān)系。JEG-3細(xì)胞中,1μmol/L和10μmol/L DAPT作用組中Notch1 mRNA表達(dá)下調(diào),兩組比較差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05);JAR細(xì)胞中,10μmol/L和15μmol/L DAPT作用組中Notch1 mRNA表達(dá)下調(diào),兩組比較差異有統(tǒng)計(jì)學(xué)意義(P0.05)。DAPT能上調(diào)JAR細(xì)胞中E-cad表達(dá)。結(jié)論:Notch信號(hào)通路和EMT與妊娠滋養(yǎng)細(xì)胞腫瘤的發(fā)生發(fā)展有關(guān);γ-分泌酶抑制劑DAPT或可成為妊娠滋養(yǎng)細(xì)胞腫瘤治療的一個(gè)新靶點(diǎn)。
[Abstract]:Aim: to investigate the important role of Notch signaling pathway in the development of human gestational trophoblastic tumor and its relationship with EMT. Methods: the proliferation of two kinds of human choriocarcinoma cell lines (JAR and JEG-3) was detected by DAPT, a 緯 -secretory enzyme inhibitor. The expression of Notch1 mRNA was detected by q-PCR and EMT in JAR cells after DAPT blocked Notch signaling pathway was detected by Western blot. Expression of related protein E-cad. Results the growth of JEG-3,JAR cells was inhibited by 10 渭 mol/L and 15 渭 mol/L DAPT for 48 h, respectively (P0.05). The inhibitory effect was in a time-and concentration-dependent manner. Notch1 mRNA expression was down-regulated in JEG-3 cells treated with 1 渭 mol/L and 10 渭 mol/L DAPT, respectively. There was no significant difference between the two groups (P0.05) the expression of Notch1 mRNA was down-regulated in the group treated with 10 渭 mol/L and 15 渭 mol/L DAPT in jar cells, and the difference between the two groups was statistically significant (P0.05) .DAPT could up-regulate the expression of E-cad in JAR cells. Conclusion the EMT and the signal pathway of Notch are related to the development of gestational trophoblastic tumors, and 緯 -secretase inhibitor DAPT may be a new target for the treatment of gestational trophoblastic tumors.
