【摘要】：目的:探究葛根素對妊娠期糖尿病大鼠胰腺組織let-7f及免疫因子IL-17、IL-23表達(dá)的影響。方法:將120只大鼠分為正常對照組、空白對照組、辛伐他汀組、葛根素低劑量組、葛根素中劑量組、葛根素高劑量組,正常對照組以基礎(chǔ)飼料、其他組以高脂飼料飼喂2周。辛伐他汀組給予2 mg/kg的辛伐他汀,葛根素組按低、中、高劑量分別給予70 mg/kg、120mg/kg、170 mg/kg葛根素,正常對照組和空白對照組給予等量生理鹽水,治療2周后,測定6組大鼠血糖和血脂水平、胰腺組織中l(wèi)et-7f的表達(dá)水平以及血清和胰腺組織中IL-17和IL-23的表達(dá)情況。結(jié)果:治療后,辛伐他汀組和葛根素組大鼠的FBG、TC、TG、FFA顯著低于空白對照組,且隨著葛根素劑量的增加,FBG、TC、TG、FFA的水平顯著降低(P0.05),呈劑量依賴性;辛伐他汀組和葛根素組胰腺組織中l(wèi)et-7f的表達(dá)顯著高于空白對照組(P0.05),且呈劑量依賴性;辛伐他汀組和葛根素組血清和胰腺組織中IL-17和IL-23的表達(dá)顯著高于空白對照組(P0.05),且呈劑量依賴性。結(jié)論:葛根素能提高妊娠期糖尿病大鼠胰腺組織let-7f表達(dá),同時降低血清和胰腺組織中IL-17、IL-23表達(dá),為妊娠期糖尿病臨床治療提供新的思路和理論依據(jù)。
[Abstract]:Aim: to investigate the effects of puerarin on the expression of let-7f and IL-17 IL-23 in pancreatic tissue of gestational diabetic rats. Methods: 120 rats were divided into normal control group, blank control group, simvastatin group, puerarin low dose group, puerarin medium dose group, puerarin high dose group, the normal control group was fed with basic diet, the other groups were fed with high fat diet for 2 weeks. Simvastatin group was given simvastatin for 2 mg/kg, puerarin group was given 70 mg / kg 120 mg / kg puerarin at low, middle and high doses, and normal control group and blank control group were given the same amount of normal saline after 2 weeks of treatment. The levels of blood glucose and blood lipid, the expression of let-7f in pancreatic tissue and the expression of IL-17 and IL-23 in serum and pancreatic tissue were measured in 6 groups. Results: after treatment, the FFA levels in simvastatin group and puerarin group were significantly lower than those in control group. The expression of let-7f in pancreatic tissue of simvastatin group and puerarin group was significantly higher than that of blank control group (P0.05), and the expression was dose-dependent. The expression of IL-17 and IL-23 in serum and pancreas in simvastatin group and puerarin group were significantly higher than those in control group (P0.05). Conclusion: puerarin can increase the expression of let-7f in pancreatic tissue of gestational diabetic rats and decrease the expression of IL-17 IL-23 in serum and pancreatic tissue, which provides a new idea and theoretical basis for the clinical treatment of gestational diabetes mellitus.
