【摘要】：目的觀察胎膜早破(PROM)患者胎膜組織Toll樣受體2(TLR2)、沉默配型信息調(diào)節(jié)蛋白6(SIRT6)及基質(zhì)金屬蛋白酶9(MMP-9)蛋白的表達變化,并探討三者間的相關(guān)性。方法選擇足月胎膜早破(t PROM)患者20例(t PROM組),未足月胎膜早破(p PROM)患者20例(p PROM組),正常晚期妊娠婦女20例(正常對照組),免疫組化法檢測各組胎膜組織TLR2、SIRT6與MMP-9表達部位,Western blotting法檢測各組胎膜組織TLR2、SIRT6與MMP-9蛋白表達水平,并分析三者的相關(guān)性。結(jié)果與正常對照組比較,t PROM及p PROM組胎膜組織中TLR2及MMP-9的表達高(P均0.01),SIRT6的表達低(P0.01),三者在t PROM組與p PROM組的表達比較,P均0.05。在胎膜組織中,TLR2與MMP-9的表達呈正相關(guān)(r=0.64,P0.05),SIRT6與MMP-9的表達呈負相關(guān)(r=-0.55,P0.05)。結(jié)論 PROM患者中,TLR2及MMP-9表達水平升高、SIRT6表達水平降低,TLR2與MMP-9的表達正相關(guān),SIRT6與MMP-9的表達負相關(guān)。
[Abstract]:Objective to investigate the expression of Toll like receptor 2 (TLR2), silent type information regulatory protein 6 (SIRT6) and matrix metalloproteinase 9 (MMP-9) in fetal membranes of patients with premature rupture of membranes (PROM). Methods Twenty (t PROM patients with term premature rupture of membranes (t PROM group), 20 patients with (p PROM) without premature rupture of membranes (p PROM group) and 20 women with normal late pregnancy (normal control group) were selected. TLR2SI-SIRT6 and MMP-9 were detected by immunohistochemical method in each group. The expression of TLR2SI-SIRT6 and MMP-9 protein were detected by Western blotting. The correlation among them is analyzed. Results compared with the normal control group, the expression of TLR2 and MMP-9 in the fetal membranes of t PROM and p PROM groups was higher (P0.01), and the expression of SIRT6 was lower (P0.01). The expression of TLR2 and MMP-9 in t PROM group and p PROM group was significantly lower than that in t PROM group and p PROM group (P 0.05). There was a positive correlation between the expression of TLR2 and MMP-9 in fetal membrane tissue (P 0.05). The expression of SIRT6 was negatively correlated with the expression of MMP-9 (P 0.05). Conclusion the expression of TLR2 and MMP-9 in patients with PROM is increased and the expression of SIRT6 is decreased. The expression of TLR2 is positively correlated with the expression of MMP-9. There is a negative correlation between the expression of SIRT6 and MMP-9.
