【摘要】：目的探討雙胎妊娠母體血中胎兒游離DNA的比例及其與妊娠周數(shù)和孕婦體重指數(shù)的關(guān)系。方法研究對(duì)象為2015-10-01—2016-08-31在中國(guó)醫(yī)科大學(xué)附屬盛京醫(yī)院產(chǎn)科就診,自愿接受無(wú)創(chuàng)產(chǎn)前檢測(cè)的妊娠11~30周雙胎孕婦。通過后續(xù)隨訪,確認(rèn)已有妊娠結(jié)局的男-男雙胎71例,其中單絨毛膜雙羊膜囊雙胎31例,雙絨毛膜雙羊膜囊雙胎40例。應(yīng)用高通量測(cè)序技術(shù),根據(jù)Y染色體的濃度估算胎兒游離DNA占孕婦血漿中總DNA的比例。結(jié)果雙胎妊娠孕婦血漿中胎兒游離DNA比例5%~30%,并與孕周呈正相關(guān)(相關(guān)系數(shù)r=0.71),與孕婦體重指數(shù)呈負(fù)相關(guān)(相關(guān)系數(shù)r=-0.78)。結(jié)論在11~30周雙胎妊娠母體血漿中,胎兒游離DNA的比例占總DNA的5%~30%,并隨著孕周的增加而上升,隨著孕婦的體重指數(shù)增加而下降。
[Abstract]:Objective to investigate the ratio of fetal free DNA in maternal blood of twin pregnancy and its relationship with gestational weeks and maternal body mass index (BMI). Methods the subjects of the study were pregnant women with 1130 weeks of gestation who voluntarily received non-invasive prenatal examination at Shengjing Hospital affiliated to China Medical University on May 10-01 to 2016-08-31. After follow-up, 71 cases of male twins with pregnancy outcome were confirmed, including 31 cases of single chorionic double amniotic sac twins and 40 cases of double chorionic double amniotic sac twins. High throughput sequencing technique was used to estimate the ratio of fetal free DNA to total DNA in maternal plasma according to the concentration of Y chromosome. Results the ratio of fetal free DNA in plasma of twin pregnant women was 30%, which was positively correlated with gestational weeks (r = 0.71) and negatively correlated with maternal body mass index (r = 0.78). Conclusion in the maternal plasma of 1130 weeks twin pregnancy, the ratio of fetal free DNA to total DNA is 5% and 30%, and increases with the increase of gestational week, and decreases with the increase of body mass index of pregnant women.
