ALDH1作為卵巢癌干細(xì)胞標(biāo)志物的實(shí)驗(yàn)研究

【摘要】:目的探討含生長(zhǎng)因子無(wú)血清培養(yǎng)基(SFM)培養(yǎng)的卵巢癌細(xì)胞株(SKOV3)細(xì)胞懸浮球?qū)D133與乙醛脫氫酶1(ALDH1)表達(dá)的影響以及ALDH1是否可作為卵巢癌干細(xì)胞(CSC)的標(biāo)志物。方法用含生長(zhǎng)因子的無(wú)血清培養(yǎng)基對(duì)卵巢癌SKOV3細(xì)胞進(jìn)行培養(yǎng)作為實(shí)驗(yàn)組,以含血清培養(yǎng)基(SSM)組作為對(duì)照,四甲基偶氮唑藍(lán)比色法(MTT)及平板克隆形成實(shí)驗(yàn)、Transwell侵襲實(shí)驗(yàn)檢測(cè)兩組細(xì)胞的自我增殖、克隆形成及侵襲能力,通過(guò)流式細(xì)胞技術(shù)檢測(cè)卵巢癌SKOV3細(xì)胞和懸浮球中ALDH1、CD133表達(dá)情況;體外化療實(shí)驗(yàn)富集ALDH1high細(xì)胞初步探索ALDH1在卵巢癌細(xì)胞中的作用。結(jié)果實(shí)驗(yàn)組自我增殖、克隆形成及侵襲能力明顯高于對(duì)照組(P0.05);實(shí)驗(yàn)組ALDH1high、CD133+細(xì)胞比例明顯高于對(duì)照組(P0.05);體外化療實(shí)驗(yàn)表明順鉑連續(xù)作用于卵巢癌SKOV3細(xì)胞72 h后,實(shí)驗(yàn)組ALDH1high、CD133+細(xì)胞比例明顯增加,ALDH1high細(xì)胞比例增加了3.23倍,CD133+細(xì)胞比例增加了2.34倍,與對(duì)照組相比較差異顯著(P0.05)。結(jié)論含生長(zhǎng)因子的無(wú)血清培養(yǎng)基培養(yǎng)卵巢癌SKOV3細(xì)胞能夠形成腫瘤干細(xì)胞球及高表達(dá)ALDH1,ALDH1可能是卵巢癌干細(xì)胞的標(biāo)志物之一。
[Abstract]:Objective to investigate the effect of suspension ball on CD133 and ALDH1 expression in ovarian cancer cell line (SKOV3) cultured on serum free medium (SFM) and whether ALDH1 can be used as a marker of ovarian cancer stem cells (CSC). Methods Ovarian cancer SKOV3 cells were cultured in serum-free medium containing growth factor as experimental group and as control group. MTT assay and plate clone formation assay were used to detect the self-proliferation, clone formation and invasion ability of the two groups, and the expression of ALDH1mCD133 in ovarian cancer SKOV3 cells and suspension spheres was detected by flow cytometry. To explore the role of ALDH1 in ovarian cancer cells by chemotherapeutic assay in vitro. Results the self-proliferation, clone formation and invasion ability of the experimental group were significantly higher than those of the control group (P0.05), the percentage of ALDH1-highCD133 cells in the experimental group was significantly higher than that in the control group (P0.05), and the chemotherapeutic experiments in vitro showed that cisplatin continued to act on ovarian cancer SKOV3 cells for 72 hours. The proportion of ALDH1 high CD133 cells in the experimental group was significantly increased by 3.23 times and the proportion of CD133 cells was increased by 2.34 times compared with the control group (P0.05). Conclusion cultured ovarian cancer SKOV3 cells in serum-free medium containing growth factor can form tumor stem cell spheres and overexpression ALDH1-ALDH1 may be one of the markers of ovarian cancer stem cells.
