MACC1在宮頸鱗癌組織中的表達(dá)及意義

【摘要】:目的:探討結(jié)腸癌轉(zhuǎn)移相關(guān)基因1(MACC1)蛋白及基因在宮頸鱗癌組織中的表達(dá),及其與宮頸鱗癌臨床分期、淋巴結(jié)轉(zhuǎn)移及分化程度的關(guān)系。 方法:采用免疫組織化學(xué)法(IHC)和半定量逆轉(zhuǎn)錄-聚合酶鏈反應(yīng)法(RT-PCR),檢測(cè)30例宮頸鱗癌組織和15例正常宮頸組織中MACC1蛋白及基因的表達(dá),并對(duì)MACC1的表達(dá)與宮頸鱗癌臨床分期、淋巴結(jié)轉(zhuǎn)移及分化程度的關(guān)系進(jìn)行統(tǒng)計(jì)學(xué)分析。 結(jié)果:1.免疫組織化學(xué)檢測(cè)結(jié)果顯示MACC1蛋白主要定位于細(xì)胞漿,在宮頸鱗癌組織中的陽(yáng)性表達(dá)率為66.7%,高于正常宮頸組織中的陽(yáng)性表達(dá)率6.67%,兩者比較,P0.05,差異具有統(tǒng)計(jì)學(xué)意義。且與宮頸鱗癌臨床分期、淋巴結(jié)轉(zhuǎn)移及分化程度相關(guān)(P0.05)。2.半定量逆轉(zhuǎn)錄-聚合酶鏈反應(yīng)法檢測(cè)結(jié)果:MACC1mRNA在宮頸鱗癌組織中的相對(duì)表達(dá)量為:0.674±0.286,高于正常宮頸組織中的相對(duì)表達(dá)量0.252±0.034,且與宮頸鱗癌臨床分期、淋巴結(jié)轉(zhuǎn)移及分化程度相關(guān)(P0.05)。 結(jié)論:MACC1表達(dá)上調(diào)可能與宮頸鱗癌(I、II期)的發(fā)展、淋巴結(jié)轉(zhuǎn)移及分化程度相關(guān)。
[Abstract]:Objective: to investigate the expression of metastasis associated gene 1 (MACC1) protein and gene in cervical squamous cell carcinoma (SCC), and its relationship with clinical stage, lymph node metastasis and differentiation of cervical squamous cell carcinoma (SCC). Methods: immunohistochemical method (IHC) and semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) were used to detect the expression of MACC1 protein and gene in 30 cases of cervical squamous cell carcinoma and 15 cases of normal cervical tissues. The relationship between lymph node metastasis and differentiation was analyzed statistically. The result is 1: 1. The results of immunohistochemistry showed that MACC1 protein was mainly located in cytoplasm, and the positive expression rate of MACC1 protein in cervical squamous cell carcinoma was 66.7%, which was higher than that in normal cervical tissue (6.67%). The difference was statistically significant (P 0.05). And it was related to the clinical stage, lymph node metastasis and differentiation of cervical squamous cell carcinoma (P0.05). The results of semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) showed that the relative expression level of WMACC1 mRNA in cervical squamous cell carcinoma was 0.674 鹵0.286, which was higher than that in normal cervical tissue (0.252 鹵0.034), and was related to the clinical stage, lymph node metastasis and differentiation of cervical squamous cell carcinoma (P0.05). Conclusion the up-regulation of MCC1 expression may be related to the development of cervical squamous cell carcinoma (stage II), lymph node metastasis and differentiation.
