【摘要】:目的:本文旨在通過研究速激肽human hemokinin-1碳端類似物hHK-1(4-11)和氮端類似物hHK-1(1-7)、hHK-1(1-8)、hHK-1(1-9)、hHK-1(1-10)對(duì)精子活力的影響,通過這種方法找出速激肽作用于精子活力的最有效片段,通過這樣的研究希望能為提高精子活力的藥物提供新的思路,為臨床的體外受精-胚胎移植以及卵胞漿內(nèi)單精子注射等輔助生殖治療手段中保持精子活力的藥物提供新的線索。方法:通過化學(xué)合成法——多肽固相合成方法合成并驗(yàn)證了5個(gè)不同的人速激肽的片段,它們分別是人速激肽的碳端片段hHK-1(4-11),以及人速激肽的氮端類似物hHK-1(1-7)、hHK-1(1-8)、hHK-1(1-9)和hHK-1(1-10)。首先,我們分別檢測(cè)了hHK-1(4-11)、hHK-1(1-7)、hHK-1(1-8)、hHK-1(1-9)和hHK-1(1-10)對(duì)正常精液PR級(jí)和PR+NP級(jí)精子活力影響,其次,我們通過流式細(xì)胞儀分析了正常成熟精子與上述藥物共培養(yǎng)3小時(shí)和6小時(shí)后精子DNA碎片的產(chǎn)生情況。結(jié)果:不同濃度(1×10-3mol/L、1×10-4mol/L、1×10-5mol/L)的hHK-1(4-11)、hHK-1(1-7)、hHK-1(1-8)、hHK-1(1-9)和hHK-1(1-10)分別作用于精子,hHK-1(1-7)作用于正常精子90min后的PR和PR+NP級(jí)精子百分率和空白對(duì)照}D比起初始的PR和PR+NP級(jí)精子百分率,精子百分率的降低具有統(tǒng)計(jì)學(xué)意義(P0.05),但是hHK-1(4-11)、hHK-1(1-8)、hHK-1(1-9)和hHK-1(1-10)作用于正常精子90min的PR和PR+NP級(jí)精子百分率的降低無統(tǒng)計(jì)學(xué)差異。結(jié)論:hHK-1(4-11)、hHK-1(1-8)、hHK-1(1-9)和hHK-1(1-10)具有保持精子活力的作用,而hHK-1(1-7)并不具備保持精子活力的作用。另一方面,通過精子DNA碎片的分析結(jié)果顯示:所有的速激肽類似物均沒有使精子DNA碎片率上升,這就說明,這些藥物并不存在精子毒性作用,其本身并不會(huì)對(duì)精子活力及完整性產(chǎn)生影響。
[Abstract]:Aim: to study the effects of fast kinin human hemokinin-1 carbon terminal analogue hHK-1 (4-11) and nitrogen terminal analogue hHK-1 (1-7) hHK-1 (1-8) hHK-1 (1-9) hHK-1 (1-10) on sperm motility, and to find out the most effective fragment of rapid kallikrein acting on sperm motility by this method. It is hoped that this study can provide new ideas for improving sperm motility and provide new clues for clinical in vitro fertilization-embryo transfer and assisted reproductive therapy such as intracytoplasmic sperm injection. Methods: five different fragments of human tachykinin, hHK-1 (4-11), hHK-1 (1-7) hHK-1 (1-8) hHK-1 (1-9) and hHK-1 (1-10), were synthesized and verified by the method of chemical synthesis and solid phase synthesis of peptides, which are the carbon terminal fragments of human tachykinin (hHK-1 (4-11) and the nitrogen terminal analogues of human tachykinin (hHK-1 (1-7) hHK-1 (1-8) hHK-1 (1-9) and hHK-1 (1-10). Firstly, we examined the effects of hHK-1 (4-11) hHK-1 (1-7) hHK-1 (1-8) hHK-1 (1-9) and hHK-1 (1-10) on the motility of normal semen PR and PR NP, respectively. The DNA fragments of normal mature spermatozoa co-cultured with these drugs for 3 and 6 hours were analyzed by flow cytometry. Results: hHK-1 (4-11) hHK-1 (1-7) hHK-1 (1-8) hHK-1 (1-9) and hHK-1 (1-10) treated with hHK-1 (1-7) and hHK-1 (1-10) at different concentrations (1 脳 10 ~ (-3) mol / L ~ (1) 脳 10 ~ (-4) mol / L ~ (-1) 脳 10 ~ (-5) mol / L) acted on the percentage of PR and PR NP grade sperm after normal sperm 90min, respectively. The decrease of sperm percentage was statistically significant (P0.05), but there was no significant difference between hHK-1 (4-11) hHK-1 (1-8) hHK-1 (1-9) and hHK-1 (1-10) on PR and PR NP level sperm percentage of normal sperm 90min. Conclusion 1: hHK-1 (4-11) hHK-1 (1-8) hHK-1 (1-9) and hHK-1 (1-10) can maintain sperm motility, but hHK-1 (1-7) does not. On the other hand, analysis of sperm DNA fragments showed that none of the tachykinin analogues increased the rate of sperm DNA fragments, suggesting that these drugs had no sperm toxicity. It does not in itself affect sperm motility and integrity.
