【摘要】：目的探索子宮內(nèi)膜異位癥盆腔粘連大鼠模型的建立方法及評價(jià)指標(biāo)。方法采用自體移植法將子宮內(nèi)膜移植于大鼠腸系膜間,術(shù)后1、3、5、7、14、21、28 d分別隨機(jī)抽取8只大鼠開腹觀察病灶生長情況,采用美國生殖學(xué)會粘連分級(AFS)分級、Haber粘連評分兩種方法同時(shí)對盆腔粘連程度進(jìn)行評分;同時(shí)采取各組病灶及周圍粘連腹膜組織行HE染色;術(shù)后5、7、14、21、28 d組分別采取病灶周圍粘連組織,對其組織型纖溶酶原激活劑(t PA)含量進(jìn)行動態(tài)檢測。結(jié)果兩種粘連評分方法均顯示造模后5 d已形成典型的盆腔粘連;與空白對照組及假手術(shù)相比,造模5 d后各模型組大鼠盆腔粘連腹膜組織中t PA含量極度降低(P0.01),造模28 d后t PA含量有上升趨勢,但仍顯著低于空白對照組及假手術(shù)組(P0.01)。結(jié)論采用自體移植法將子宮內(nèi)膜移植于腸系膜間建立EMs盆腔粘連模型,造模術(shù)后5 d即為成熟模型,t PA與EMs盆腔粘連的發(fā)生具有相關(guān)性。
[Abstract]:Objective to explore the method and evaluation index of pelvic adhesion model of endometriosis in rats. Methods the endometrium was transplanted into mesenteric mesentery of rats by autologous transplantation. Eight rats were randomly selected to observe the growth of the mesentery on the 28th day after operation. The degree of pelvic adhesions was evaluated by the American Society of Reproduction Association (AFS) grade and Haber's adhesion grade, and HE staining was used in each group of lesions and surrounding adhesion peritoneum tissue at the same time. The tissue type plasminogen activator (t PA) content was measured dynamically in the postoperative group of 28 days after operation with adherent tissue around the lesion. Results the two adhesion scoring methods showed that typical pelvic adhesions had been formed 5 days after modeling, compared with blank control group and sham operation. After 5 days, the content of t-PA in the peritoneal tissue of the rats with pelvic adhesions was extremely decreased (P0.01), and the content of t-PA increased after 28 days, but still significantly lower than that of the blank control group and sham-operation group (P0.01). Conclusion the pelvic adhesion model of endometrium was established by autologous transplantation of endometrium into mesentery. The model is mature model 5 days after operation. There is a correlation between t PA and pelvic adhesion.
