殼聚糖凝膠用于避孕的機制及可行性研究

【摘要】:目的研究殼聚糖凝膠對精子活力的影響及其影響機制并初步探討殼聚糖凝膠用于避孕的可行性。方法將200例精液常規(guī)在正常范圍的精液標本隨機分為兩組,每組各100例,對照組無任何處理,殼聚糖凝膠組加入殼聚糖凝膠。分別測定并比較兩組中活動力為a級(快速向前運動)與b級(慢速或呆滯的前向運動)的精子百分率;形態(tài)學正常、頭部缺陷、體部缺陷及尾部缺陷的精子數(shù);精子頂體酶活性的定量值;精漿酸性磷酸酶、中性α-葡糖苷酶及鋅的含量。結果殼聚糖凝膠組精子活動力較對照組顯著降低,兩組活動力為a級的精子百分率差異有統(tǒng)計學意義(P0.05),活動力為b級的精子百分率差異無統(tǒng)計學意義(P0.05)。殼聚糖凝膠組畸形精子數(shù)較對照組顯著增加,兩組形態(tài)學正常、頭部缺陷尤其是尾部缺陷的精子數(shù)差異均有統(tǒng)計學意義(P0.05),而中段缺陷的精子數(shù)差異無統(tǒng)計學意義(P0.05)。殼聚糖凝膠組精子頂體酶活性的定量值較對照組降低,但差異無統(tǒng)計學意義(P0.05)。殼聚糖凝膠組精漿酸性磷酸酶、中性α-葡糖苷酶及鋅的含量均較對照組明顯降低,差異均有統(tǒng)計學意義(P0.05)。結論殼聚糖凝膠可顯著降低精子的活動力并致精子畸形,同時引起精漿各生化指標不同程度地降低,因而具有顯著的體外殺精作用,值得進一步研究、開發(fā)為一種新型的殺精劑類外用避孕藥物。
[Abstract]:Objective to study the effect of chitosan gel on sperm motility and its mechanism and to explore the feasibility of using chitosan gel for contraception. Methods 200 semen samples were randomly divided into two groups, 100 cases in each group. The control group had no treatment, and the chitosan gel group was added with chitosan gel. The percentage of spermatozoa in the two groups were measured and compared, the sperm numbers of normal morphology, head defect, body defect and tail defect were measured and compared between the two groups. The quantitative value of acrosin activity in spermatozoa, the content of acid phosphatase, neutral 偽 -glucosidase and zinc in seminal plasma. Results the sperm motility of the chitosan gel group was significantly lower than that of the control group. There was a significant difference in the percentage of spermatozoa with a grade a between the two groups, but there was no significant difference in the percentage of the spermatozoa with the b grade in the two groups. The number of abnormal sperm in chitosan gel group was significantly higher than that in control group. The morphology of the two groups was normal. The difference of sperm number in head defect, especially in tail defect was statistically significant (P 0.05), but there was no significant difference in sperm count in middle segment (P 0.05). The quantitative value of acrosin activity in chitosan gel group was lower than that in control group, but the difference was not statistically significant (P 0.05). The contents of acid phosphatase, neutral 偽 -glucosidase and zinc in the seminal plasma of chitosan gel group were significantly lower than those in the control group (P 0.05). Conclusion chitosan gel can significantly reduce the motility of sperm and cause sperm deformity, at the same time, the biochemical indexes of seminal plasma can be decreased to some extent, so it has remarkable spermicidal effect in vitro, which is worthy of further study. To develop a new spermicide for external use.
