【摘要】：目的了解SFRP2在人宮頸癌組織中的表達變化,探討SFRP2對宮頸癌細胞增殖的影響。方法采用Western bolt及qRT-PCR檢測SFRP2在宮頸癌組織中的表達;用慢病毒構(gòu)建過表達SFRP2的人宮頸癌細胞株,并用CCK-8及平板克隆分析SFRP2對細胞增殖的影響;Western bolt和qRT-PCR檢測SFRP2在腫瘤細胞內(nèi)對WNT通路的相關(guān)蛋白和基因表達的影響。結(jié)果與癌旁組織相比,SFRP2在人宮頸癌組織中低表達;過表達SFRP2的宮頸癌細增殖受抑制;SFRP2抑制細胞增殖是通過WNT信號通路而發(fā)生。結(jié)論 SFRP2作為宮頸癌新的候選基因作用有待深入研究。
[Abstract]:Objective to investigate the expression of SFRP2 in human cervical carcinoma and to investigate the effect of SFRP2 on the proliferation of cervical cancer cells. Methods the expression of SFRP2 in cervical carcinoma tissue was detected by Western bolt and qRT-PCR, and the human cervical cancer cell line expressing SFRP2 was constructed by lentivirus. The effects of SFRP2 on cell proliferation were analyzed by CCK-8 and plate cloning. The effects of SFRP2 on the expression of WNT related proteins and genes in tumor cells were detected by Western bolt and qRT-PCR. Results the expression of SFRP2 in human cervical carcinoma tissues was lower than that in paracancerous tissues, and the inhibition of fine proliferation of cervical carcinoma cells with overexpression of SFRP2 was mediated by WNT signaling pathway. Conclusion the role of SFRP2 as a new candidate gene for cervical cancer needs further study.
