CCCTC-結(jié)合因子(CTCF)及端粒酶hTERT在人子宮內(nèi)膜癌組織中的表達(dá)研究

【摘要】:第一部分應(yīng)用Western-blotting技術(shù)檢測人子宮內(nèi)膜癌組織中CTCF和端粒酶基因hTERT蛋白表達(dá)的研究 目的:檢測對比人子宮內(nèi)膜癌和正常子宮內(nèi)膜組織CTCF和端粒酶基因hTERT蛋白表達(dá)情況,結(jié)合臨床病理參數(shù)分析子宮內(nèi)膜癌組CTCF及hTERT分別與之關(guān)聯(lián),最后分析CTCF與hTERT之間聯(lián)系,為臨床奠定一定基礎(chǔ)。 方法:隨機(jī)選取30例正常人子宮內(nèi)膜組織標(biāo)本(其中15例增殖期內(nèi)膜組織,15例分泌期內(nèi)膜組織)為對照組,45例子宮內(nèi)膜癌患者癌組織標(biāo)本為實(shí)驗(yàn)組。采用Western-blotting法檢測CTCF蛋白和端粒酶基因hTERT(Human telomerase reverse transcriptase,hTERT)蛋白在各組的表達(dá)水平。 結(jié)果:CTCF和hTERT在各組中均有表達(dá)。實(shí)驗(yàn)組CTCF蛋白和端粒酶基因hTERT蛋白陽性表達(dá)率均高于對照組。實(shí)驗(yàn)組與對照組間CTCF和端粒酶基因hTERT蛋白相對表達(dá)量均存在統(tǒng)計(jì)學(xué)差異。子宮內(nèi)膜癌組CTCF表達(dá)與子宮內(nèi)膜癌的病理分級、臨床分期均有關(guān),與病理類型無關(guān);實(shí)驗(yàn)組hTERT表達(dá)與子宮內(nèi)膜癌病理分級、病理類型、臨床分期均無關(guān)。子宮內(nèi)膜癌組CTCF與hTERT異常的表達(dá)存在正相關(guān)性。 結(jié)論:CTCF與端粒酶基因hTERT蛋白在人子宮內(nèi)膜癌和正常子宮內(nèi)膜中表達(dá)存在差異性;不同臨床病理參數(shù)下CTCF和hTERT表達(dá)情況各異;在子宮內(nèi)膜癌中兩者聯(lián)合檢測對子宮內(nèi)膜癌的診斷具有一定的意義。 第二部分應(yīng)用半定量RT-PCR法檢測人子宮內(nèi)膜癌組織中CTCF和hTERTmRNA表達(dá)的研究 目的:檢測對比人子宮內(nèi)膜癌組織與正常子宮內(nèi)膜組織中CTCFmRNA與hTERTmRNA的表達(dá)情況,分析其臨床意義。 方法:從收集好的30例人正常子宮內(nèi)膜標(biāo)本(15例增殖期內(nèi)膜組織,15例分泌期內(nèi)膜組織)中挑選出WB實(shí)驗(yàn)CTCF表達(dá)陽性樣本組織進(jìn)行CTCFmRNA檢測、hTERT表達(dá)陽性樣本組織進(jìn)行hTERTmRNA檢測;從45例子宮內(nèi)膜癌患者的癌組織標(biāo)本中挑選出WB實(shí)驗(yàn)CTCF表達(dá)陽性樣本組織進(jìn)行CTCFmRNA檢測、hTERT表達(dá)陽性樣本組織進(jìn)行hTERTmRNA檢測;分別先后提取RNA,采用半定量RT-PCR法檢測各標(biāo)本中mRNA表達(dá)情況。 結(jié)果:子宮內(nèi)膜癌組CTCFmRNA和端粒酶hTERTmRNA各自相對表達(dá)量與對照組間存在統(tǒng)計(jì)學(xué)差異。 結(jié)論:CTCFmRNA和端粒酶hTERTmRNA各自在實(shí)驗(yàn)組與對照組間存在不同,利用這一差異性可為臨床診斷及治療提供一定的基礎(chǔ)。
[Abstract]:The first part used Western-blotting technique to detect the expression of CTCF and telomerase gene hTERT protein in human endometrial carcinoma. Objective: to detect and compare the expression of CTCF and telomerase gene hTERT protein in human endometrial carcinoma and normal endometrial tissue. The relationship between CTCF and hTERT in endometrial carcinoma was analyzed with clinicopathological parameters. Finally, the relationship between CTCF and hTERT was analyzed, which laid a foundation for clinical practice. Methods: 30 normal human endometrial tissues (15 proliferative endometrial tissues and 15 secretory endometrial tissues) were randomly selected as control group and 45 endometrial carcinoma specimens as experimental group. Western blotting was used to detect the expression of CTCF protein and human telomerase reverse transcriptasehTERT protein in each group. Results the expression of 1% CTCF and hTERT was found in all groups. The positive expression rate of CTCF protein and telomerase gene hTERT protein in experimental group was higher than that in control group. The relative expression of CTCF and telomerase gene hTERT was significantly different between the experimental group and the control group. The expression of CTCF in endometrial carcinoma group was related to the pathological grade and clinical stage of endometrial carcinoma, but not to pathological type, while hTERT expression in experimental group was not related to pathological grade, pathological type and clinical stage of endometrial carcinoma. There was a positive correlation between CTCF and hTERT expression in endometrial carcinoma. Conclusion there is a difference between the expression of human CTCF and telomerase gene hTERT in human endometrial carcinoma and normal endometrium, the expression of CTCF and hTERT is different under different clinicopathologic parameters. The combined detection of both in endometrial carcinoma has a certain significance in the diagnosis of endometrial carcinoma. The second part used semi-quantitative RT-PCR to detect the expression of CTCF mRNA and hTERT mRNA in human endometrial carcinoma. Objective: to detect and compare the expression of CTCF mRNA and hTERT mRNA in human endometrial carcinoma and normal endometrial tissue, and to analyze its clinical significance. Methods: a total of 15 cases of proliferative endometrial tissues were collected from 30 normal human endometrium specimens. The samples of CTCF positive expression were collected from 15 cases of secretory endometrial tissues in proliferative phase. The CTCF mRNA positive samples were detected for hTERT mRNA detection. From 45 samples of endometrial carcinoma, the samples of CTCF positive expression were collected from 45 patients with endometrial carcinoma. The positive samples of CTCF mRNA and hTERT expression were detected for hTERT mRNA. RNA was extracted successively and mRNA expression was detected by semi-quantitative RT-PCR. Results: the relative expressions of CTCF mRNA and telomerase hTERT mRNA in endometrial carcinoma group were significantly different from those in control group. Conclusion there are differences between the experimental group and the control group in the two groups. The use of this difference can provide a basis for clinical diagnosis and treatment.
