卵巢上皮癌組織中hMSH2的表達(dá)及其與化療耐藥的關(guān)系

【摘要】:目的檢測(cè)不同化療敏感程度的卵巢上皮癌組織中hMSH2基因的表達(dá),分析其與化療耐藥及預(yù)后的關(guān)系。方法 80例新鮮卵巢上皮癌組織均由手術(shù)中取得,采用ATP-TCA技術(shù)檢測(cè)卵巢癌組織對(duì)八種藥物:紫杉醇、卡鉑、拓泊替康、多西他賽、吉西他濱、環(huán)磷酰胺體內(nèi)代謝產(chǎn)物、依托泊苷及博來霉素的體外敏感性。采用Real-time PCR及Westernblot法分別檢測(cè)80例卵巢上皮癌組織中hMSH2基因mRNA和蛋白水平的表達(dá)情況。結(jié)果對(duì)紫杉醇耐藥的卵巢上皮癌組中,hMSH2基因在mRNA及蛋白水平的表達(dá)均明顯高于敏感組(P0.01),在對(duì)卡鉑、拓泊替康、多西他賽耐藥組中hMSH2表達(dá)水平也明顯高于敏感組(P0.05);hMSH2基因的相對(duì)表達(dá)值與卡鉑、拓泊替康、多西他賽的敏感度系數(shù)有顯著相關(guān)性(P≤0.05)。早期卵巢癌患者組織的hMSH2表達(dá)水平明顯低于晚期卵巢癌組織(P0.05)。高、中分化卵巢癌組織hMSH2蛋白表達(dá)水平明顯低于低分化卵巢癌組織(P=0.000)。原發(fā)卵巢癌患者組織的hMSH2表達(dá)水平明顯低于復(fù)發(fā)卵巢癌患者組織(P0.01)。結(jié)論 hMSH2基因的高表達(dá)與卵巢癌化療耐藥及預(yù)后相關(guān),化療可能誘導(dǎo)hMSH2基因的高表達(dá);下調(diào)hMSH2基因的表達(dá)或許是逆轉(zhuǎn)腫瘤耐藥的新途徑。
[Abstract]:Objective to detect the expression of hMSH2 gene in ovarian epithelial carcinoma with different chemosensitivity, and to analyze its relationship with chemotherapeutic resistance and prognosis. Methods Eighty cases of fresh ovarian epithelial carcinoma were obtained by operation. ATP-TCA technique was used to detect the metabolites of eight drugs in ovarian cancer tissues: paclitaxel, carboplatin, topotecan, docetaxel, gemcitabine, cyclophosphamide. In vitro sensitivity of etoposide and bleomycin. Real-time PCR and Western blot were used to detect the expression of hMSH2 mRNA and protein in 80 cases of ovarian epithelial carcinoma. Results the expression of hMSH2 gene in paclitaxel-resistant ovarian epithelial carcinoma was significantly higher than that in sensitive group (P 0.01), and in carboplatin and topotecan. The expression level of hMSH2 in docetaxel resistant group was significantly higher than that in sensitive group (P < 0.05).
