抑制URG4表達(dá)對宮頸癌小鼠的治療作用及機(jī)制

【摘要】:目的:分析上調(diào)基因-4(URG4)在宮頸癌中的表達(dá),并觀察分析抑制URG4對宮頸癌的治療作用及機(jī)制。方法:Western blot法分析URG4在宮頸癌組織及癌旁組織中的表達(dá)差異。選擇16只雄性BALB/c裸鼠,喂養(yǎng)至6周齡后隨機(jī)分為對照組(Control組)及觀察組(Observation組),每組各8只。Control組小鼠于右側(cè)背部皮下接種宮頸癌細(xì)胞(Si Ha cells)。Observation組小鼠背部皮下接種URG4-si RNA轉(zhuǎn)染的宮頸癌細(xì)胞(URG4-si RNA Si Ha cells)。干預(yù)后4周處死小鼠,分析對比小鼠腫瘤生長情況及Wnt/β-catenin信號通路的改變。結(jié)果:URG4在人宮頸癌組織中呈現(xiàn)高表達(dá),顯著高于癌旁組織(P0.05)。干預(yù)結(jié)束后,觀察組小鼠腫瘤組織內(nèi)URG4表達(dá)量顯著低于對照組(P0.05);體重明顯高于對照組(P0.05);腫瘤組織重量顯著低于對照組(P0.05)。觀察組小鼠腫瘤組織內(nèi)Ki67表達(dá)量顯著低于對照組(P0.05)。觀察組小鼠腫瘤組織內(nèi)Cyclin D1、β-catenin及C-myc的表達(dá)均顯著低于對照組,相比較差異均具有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:URG4在人宮頸癌中呈現(xiàn)高表達(dá),抑制URG4的表達(dá)可能通過抑制Wnt/β-catenin信號通路而發(fā)揮抗腫瘤效應(yīng)。
[Abstract]:Aim: to investigate the expression of upregulation gene -4 urg _ 4 in cervical cancer, and to investigate the therapeutic effect and mechanism of inhibition of URG4 on cervical cancer. Methods the expression of URG4 in cervical carcinoma and paracancerous tissues was analyzed by Western blot. Sixteen male BALB / R / c nude mice were selected. After feeding to the age of 6 weeks, mice were randomly divided into control group (n = 8) and observation group (n = 8 in each group). The mice in each group were inoculated subcutaneously with cervix carcinoma cell line (Cervical carcinoma cell line) Si Ha cells transfected with URG4-si RNA on the back of mice in the right back of the mice in the control group and in the control group. The mice in the observation group were inoculated subcutaneously with URG4-si RNA transfected with URG4-si RNA. The mice were killed 4 weeks after intervention. The tumor growth and the changes of Wnt / 尾-catenin signaling pathway were analyzed and compared. Results the expression of 1% URG4 in human cervical carcinoma was higher than that in paracancerous tissue (P 0.05). After the intervention, the expression of URG4 in tumor tissues in the observation group was significantly lower than that in the control group (P 0.05), the body weight was significantly higher than that in the control group (P 0.05), and the weight of the tumor tissue was significantly lower than that in the control group (P 0.05). The expression of Ki67 in tumor tissues in the observation group was significantly lower than that in the control group (P 0.05). The expression of Cyclin D1, 尾 -catenin and C-myc in the tumor tissues of the observation group was significantly lower than that in the control group, and the difference was statistically significant (P 0.05). Conclusion the expression of 1: URG4 is highly expressed in human cervical carcinoma. Inhibition of the expression of URG4 may play an antitumor effect by inhibiting Wnt- 尾 -catenin signaling pathway.
