輔助生殖技術(shù)對人胎盤DNA甲基化水平的影響

【摘要】:目的探究輔助生殖技術(shù)(ART)助孕對單胎足月分娩新生兒的胎盤及胎盤總DNA甲基化水平和甲基化相關(guān)基因表達的影響。方法選擇2016年6月—2016年12月期間單胎足月分娩的患者,ART子代為ART組,自然妊娠子代為對照組,兩組按孕婦年齡、孕周等進行配對后,比較兩組胎盤質(zhì)量、新生兒體質(zhì)量、胎盤效率。采用酶聯(lián)免疫吸附測定法(ELISA)測定胎盤總DNA的甲基化水平,采用實時定量聚合酶鏈式反應(RT-q PCR)方法檢測胎盤甲基轉(zhuǎn)移酶(DNA methyltransferase,DNMT)家族和去甲基化酶(ten-eleven translocation,TET)家族的m RNA表達水平。結(jié)果 (1)ART組新生兒體質(zhì)量[(3 650.50±393.84)g]、胎盤重量[(633.05±98.58)g]顯著增加,且高于對照組[(3 339.50±377.01)g,(563.00±85.60)g](P0.05)。(2)ART組胎盤總DNA甲基化水平(1.22%±0.45%)顯著低于對照組(2.08%±0.98%)(P0.05)。(3)ART組DNMT3B、TET1、TET2、TET3的m RNA相對表達量分別為4.61±1.06、4.66±1.06、4.99±1.09、4.92±1.13,顯著高于對照組(1.23±0.55、1.27±0.56、1.47±0.50、1.41±0.61)(P0.05),DNMT1、DNMT3A的表達水平無統(tǒng)計學差異(P0.05)。結(jié)論 ART會導致人類妊娠晚期胎盤過度生長,總DNA甲基化水平降低,DNMT3B、TET1、TET2、TET3表達上調(diào)可能是導致胎盤DNA甲基化水平降低的機制。
[Abstract]:Objective to investigate the effect of assisted reproductive technique (ART) on the total DNA methylation level and methylation-related gene expression in placenta and placenta of newborns with single term delivery. Methods from June 2016 to December 2016, patients with single term delivery were selected as art group and natural pregnancy as control group. The two groups were matched according to the age and gestational weeks of pregnant women, and the placental weight and body weight of newborns were compared between the two groups.
