正常宮頸、宮頸上皮內(nèi)瘤變Ⅲ級和宮頸癌的拉曼光譜分析

【摘要】:目的:(1)采用拉曼光譜儀對正常宮頸、CIN III和宮頸癌進(jìn)行光譜分析比較,總結(jié)各自的特征峰,對其進(jìn)行分析總結(jié);(2)拉曼光譜技術(shù)對宮頸癌、CIN III組織的光譜分析是否可用于臨床篩查,并成為宮頸癌及癌前病變篩查的一種新方法。 材料和方法:組織標(biāo)本取自于2013年4月—2013年11月在吉林大學(xué)第一醫(yī)院婦科行手術(shù)治療的患者,共63例。其中,宮頸鱗狀細(xì)胞癌組織標(biāo)本33例、CIN III組織標(biāo)本10例為實(shí)驗(yàn)組,正常宮頸組織標(biāo)本20例為對照組,采用拉曼光譜儀對正常宮頸組織、CIN III組織和宮頸癌組織進(jìn)行光譜分析,得出各自的拉曼圖譜,并對其分析。 結(jié)果:1、在不同組織中拉曼光譜的特征峰的位置不同,具體表現(xiàn):正常宮頸組織主要的拉曼光譜特征峰在640、853、928、1005、1242、1450、1660-1cm;CIN III組織主要的拉曼光譜特征峰在853、931、1003、1242、1341、1447、1657-1cm;宮頸癌主要的拉曼光譜特征峰在642、853、1003、1345、1443、1657-1cm。 2、拉曼光譜在不同組織中同一特征峰的相對強(qiáng)度不同:以核酸為主要特征峰的1345cm-1處,核酸含量差異較大,依次為宮頸癌組織>CIN III組織>正常宮頸組織(P<0.01);以蛋白質(zhì)為主要特征峰的1003cm-1處,,蛋白質(zhì)含量差異較大,依次為宮頸癌>CIN III組織>正常宮頸組織(P<0.01);以脂類為特征峰的1443cm-1處,正常組織的相對強(qiáng)度高于CIN III組織和宮頸癌的相對強(qiáng)度,但三者的相對強(qiáng)度無明顯差異(P>0.05)。 結(jié)論:1、拉曼光譜在正常宮頸組織、CIN III、宮頸癌組織中其特征峰的位置不同。2、拉曼光譜在正常宮頸組織、CIN III、宮頸癌組織中同一特征峰的相對強(qiáng)度不同。3、拉曼光譜技術(shù)有望成為篩查宮頸癌和宮頸癌前病變的檢測方法。
[Abstract]:Objective: to analyze and compare the spectra of cin III and cervical cancer by Raman spectrometer, summarize their characteristic peaks and sum up their characteristic peaks. Whether Raman spectroscopy can be used for clinical screening of cin III tissue from cervical cancer. It has become a new method for screening cervical cancer and precancerous lesions. Materials and methods: tissue specimens were collected from 63 patients who underwent gynecologic surgery in the first Hospital of Jilin University from April 2013 to November 2013. Among them, 33 specimens of cervical squamous cell carcinoma (SCC) and 10 specimens of cin III (n = 10) were used as experimental group and 20 cases as control group. Raman spectrometer was used to analyze cin III tissue and cervical carcinoma tissue of normal cervix. The Raman spectra were obtained and analyzed. Results the characteristic peaks of Raman spectra in different tissues were different in different tissues. The main Raman characteristic peaks of normal cervix were 640853928 / 1005 / 1242n ~ 1450 ~ 1660-1cm ~ (-1). The main characteristic peaks of Raman spectra of cin / III were 853931 / 100312441 / 14447 / 1657-1 cm and 642853313445 / 14431657-1cm / cm, respectively, and the main characteristic peaks of Raman spectra of cervical cancer were 64285313455 / 14431657-1cm respectively, and the main characteristic peaks of Raman spectra of cervical cancer were 64285313455 ~ 14431657-1cm ~ (-1) cm. 2. The relative intensity of the same characteristic peak in different tissues is different: the nucleic acid content is different at the 1345cm-1 where nucleic acid is the main characteristic peak. The order was cervical cancer > CIN III > normal cervix (P < 0.01), the content of protein in 1003cm-1 with protein as the main characteristic peak was higher than that in normal cervix (P < 0.01), and that in 1443cm-1 with lipid as the characteristic peak was higher than that in normal cervix (P < 0.01), the order was cervical cancer > CIN III > normal cervix (P < 0.01). The relative strength of normal tissue was higher than that of CIN III and cervical cancer, but there was no significant difference among them (P > 0.05). Conclusion in normal cervical tissue, the Raman spectrum has different position of characteristic peak in cervical cancer tissue, Raman spectrum in normal cervix tissue of cin III, Raman spectrum in normal cervix tissue of cin III, and the relative intensity of the same characteristic peak in cervical cancer tissue is different. It is expected to be a screening method for cervical cancer and cervical precancerous lesions.
