孕鼠鐵缺乏對(duì)子代GH-IGF-1軸的影響

【摘要】:目的探討大鼠妊娠期鐵缺乏對(duì)新生仔鼠生長(zhǎng)激素-胰島素樣生長(zhǎng)因子-1(GH-IGF-1)軸的影響及機(jī)制。方法 4周齡雌性Wistar大鼠36只,隨機(jī)分為實(shí)驗(yàn)組和對(duì)照組,分別飼低鐵飼料和含鐵飼料喂養(yǎng)4周,檢測(cè)雌鼠血紅蛋白(Hb)濃度,血細(xì)胞比容(HCT)、紅細(xì)胞平均容積(MCV)、紅細(xì)胞平均血紅蛋白量(MCH)、紅細(xì)胞平均血紅蛋白濃度(MCHC)及紅細(xì)胞分布寬度(RDW)等血液學(xué)相關(guān)參數(shù)。將雌鼠與正常雄鼠合籠,孕末期檢測(cè)血液學(xué)相關(guān)參數(shù)并行骨髓鐵染色。觀察仔鼠存活率及體重。實(shí)時(shí)熒光定量聚合酶鏈?zhǔn)椒磻?yīng)(qRTPCR)方法檢測(cè)新生仔鼠垂體生長(zhǎng)激素(GH)及肝臟胰島素樣生長(zhǎng)因子-1(IGF-1)表達(dá)量。結(jié)果相比對(duì)照組,實(shí)驗(yàn)組低鐵干預(yù)4周后孕鼠孕末期Hb、HCT、MCV、MCH及MCHC等水平降低(P0.05),RDW水平升高(P0.05);實(shí)驗(yàn)組大鼠骨髓細(xì)胞內(nèi)鐵及外鐵減少;實(shí)驗(yàn)組新生仔鼠體重及仔鼠存活率低于對(duì)照組(P0.05);實(shí)驗(yàn)組垂體GH信使核糖核酸(mRNA)及肝臟IGF-1 mRNA的表達(dá)低于對(duì)照組(P0.05)。結(jié)論妊娠期鐵缺乏下調(diào)子代GH和IGF-1 mRNA表達(dá)造成子代低出生體重。
[Abstract]:Objective to investigate the effect and mechanism of iron deficiency during pregnancy on growth hormone-insulin-like growth factor-1 (GH-IGF-1) axis in newborn rats. Methods Thirty-six 4-week-old female Wistar rats were randomly divided into experimental group and control group. Hematological parameters such as HCT, mean volume of red blood cell (MCV), mean hemoglobin content (MCHN), mean hemoglobin concentration (MCHCC) and distribution width of red blood cell (RDW) were measured. Bone marrow iron staining was performed at the end of pregnancy to detect the hematologic parameters of female mice and normal male mice. The survival rate and body weight were observed. The expression of growth hormone (GH) and insulin-like growth factor-1 (IGF-1) in liver of newborn rats were detected by real-time fluorescence quantitative polymerase chain reaction (QRT PCR). Results compared with the control group, the levels of HCT, MCV, MCH and MCHC in the late pregnant rats of the experimental group were decreased after the intervention of low iron for 4 weeks, and the levels of RDW in the bone marrow cells of the experimental group were increased, and the levels of iron and external iron in the bone marrow cells of the experimental group were decreased. The body weight and survival rate of newborn rats in the experimental group were lower than those in the control group (P 0.05), and the expression of GH mRNA mRNAs and IGF-1 mRNA in the liver in the experimental group were lower than those in the control group (P 0.05). Conclusion Iron deficiency during pregnancy can decrease the expression of GH and IGF-1 mRNA in offspring and result in low birth weight.
