Furin抑制劑α1-PDX對(duì)宮頸癌HeLa細(xì)胞生長(zhǎng)、轉(zhuǎn)移和成瘤的影響

【摘要】:目的研究α1-PDX對(duì)宮頸癌細(xì)胞株He La細(xì)胞生長(zhǎng)、轉(zhuǎn)移及成瘤的影響并探討其機(jī)制。方法α1-PDX轉(zhuǎn)染He La細(xì)胞,通過(guò)MTT細(xì)胞增殖實(shí)驗(yàn),Boyden細(xì)胞遷移和侵襲實(shí)驗(yàn)觀察He La細(xì)胞的生長(zhǎng),遷移和侵襲能力。采用蛋白印跡法檢測(cè)細(xì)胞Furin及產(chǎn)物膜性Ⅰ型金屬蛋白酶(MT1-MMP)蛋白量的變化,熒光酶底物實(shí)驗(yàn)檢測(cè)Furin活性。制作He La細(xì)胞致瘤裸鼠模型研究腫瘤體內(nèi)生長(zhǎng)。結(jié)果細(xì)胞增殖實(shí)驗(yàn)結(jié)果顯示,與對(duì)照組相比24 hα1-PDX組He La細(xì)胞體外生長(zhǎng)抑制18.4%,差異有統(tǒng)計(jì)學(xué)意義(P0.01);細(xì)胞遷移和侵襲實(shí)驗(yàn)結(jié)果顯示,α1-PDX處理組穿過(guò)濾膜和基質(zhì)膠的He La細(xì)胞數(shù)量均明顯少于對(duì)照組(P0.01);He La/PDX可顯著降低Furin的活性和MTI-MMP的蛋白水平。He La/PDX致瘤裸鼠的成瘤機(jī)率及皮下腫瘤體積明顯小于對(duì)照組。結(jié)論α1-PDX可有效抑制人宮頸癌He La細(xì)胞的生長(zhǎng)、轉(zhuǎn)移及成瘤能力,機(jī)制可能與降低Furin活性及產(chǎn)物MTI-MMP的表達(dá)有關(guān)。
[Abstract]:Objective to study the effect of 偽 1-PDX on the growth, metastasis and tumorigenesis of cervical cancer cell line He-La. Methods He-La cells were transfected with 偽 1-PDX. The growth, migration and invasion of He-La cells were observed by MTT cell proliferation assay. The changes of Furin and MT1-MMPin protein were detected by Western blot, and the activity of Furin was detected by fluorescence enzyme substrate assay. A nude mouse model of helium La cell tumorigenesis was established to study the tumor growth in vivo. Results the results of cell proliferation test showed that, Compared with the control group, the growth inhibition of He-La cells in vitro in 24 h 偽 1-PDX group was significantly lower than that in the control group (P 0.01). The results of cell migration and invasion experiments showed that the number of He-La cells in 偽 1-PDX treatment group was significantly lower than that in control group (P 0.01), and the number of He-La cells in 偽 1-PDX treatment group was significantly lower than that in the control group (P 0.01). La/PDX significantly decreased the activity of Furin and the protein level of MTI-MMP. The probability of tumorigenesis and subcutaneous tumor volume in nude mice induced by he La/PDX were significantly lower than those in the control group. Conclusion 偽 1-PDX can effectively inhibit the growth, metastasis and tumorigenesis of human cervical cancer He-La cells. The mechanism may be related to the decrease of Furin activity and the expression of MTI-MMP.
