XIAP、VEGF在子癇前期中的表達(dá)及相關(guān)性研究

【摘要】:目的:探討分析XIAP、VEGF在子癇前期組孕婦與對照組孕婦血清、胎盤組織中的表達(dá)情況,研究其對子癇前期的影響及二者的相關(guān)性,為子癇前期臨床治療及科研工作提供依據(jù)。 方法:選取30例2012年11月至2013年7月在廣西醫(yī)科大學(xué)第一附屬醫(yī)院住院孕產(chǎn)婦中,子癇前期的患者30例,其中20例輕度子癇前期患者,10例重度子癇前期患者。隨機(jī)抽選同期妊娠晚期的正常孕婦共20例做為對照組。采集病例的臨床特征資料進(jìn)行比較;采用親和免疫細(xì)胞化學(xué)技術(shù)鏈霉素-生物素-過氧化物酶復(fù)合物技術(shù)(SABC)法測定胎盤組織中XIAP、VEGF蛋白的表達(dá)。通過ELISA技術(shù)對XIAP、VEGF在正常妊娠孕婦和子癇前期孕婦外周血中的表達(dá)含量進(jìn)行測定。 結(jié)果:(1)子癇前期患者血清中XIAP、VEGF表達(dá)水平分別為(203.92士19.97)mg/L、(116.32士11.86)mg/L,對照組孕婦血清中XIAP、VEGF表達(dá)水平分別為(300.88士25.50)mg/L、(164.47士15.47)mg/L。XIAP、VEGF在子癇前期組和正常對照組血清中的表達(dá)量差異具有統(tǒng)計(jì)學(xué)意義(P均<0.05);XIAP、VEGF在輕度子癇前期組血清中的表達(dá)量分別為(215.56士9.60)mg/L、(122.81士8.24)mg/L,XIAP、VEGF在重度子癇前期組血清中的表達(dá)量分別為(180.62士13.66)mg/L、(103.32士5.35)mg/L,輕、重度子癇前期組血清中XIAP、VEGF相對表達(dá)量差異均有統(tǒng)計(jì)學(xué)意義,XIAP、VEGF在血清中的表達(dá)量呈正相關(guān)(r=0.929)。(2)XIAP在對照組、輕度和重度子癇前期組胎盤組織中的陽性表達(dá)率分別為100%、84%和62%,其中輕度子癇前期組與對照組相比,差異有統(tǒng)計(jì)學(xué)意義(χ2=17.391,P=0.000),重度子癇前期組與輕度子癇前期組相比,差異有統(tǒng)計(jì)學(xué)意義(χ2=9.019,P=0.003);VEGF在對照組、輕度和重度子癇前期組胎盤組織中的陽性表達(dá)率分別為100%、91%和72%,其中輕度患者組與對照組相比,差異有統(tǒng)計(jì)學(xué)意義(χ2=10.497,,P=0.001),重度患者組與輕度患者組相比,差異有統(tǒng)計(jì)學(xué)意義(χ2=9.269,P=0.002)。 結(jié)論: XIAP、VEGF可能參與了子癇前期的發(fā)生發(fā)展。
[Abstract]:Objective: to investigate the expression of XIAPP VEGF in serum and placenta of pregnant women with preeclampsia and control group, and to study its effect on preeclampsia and the correlation between them, and to provide the basis for clinical treatment and scientific research of preeclampsia. Methods: from November 2012 to July 2013, 30 pregnant women with pre-eclampsia were enrolled in the first affiliated Hospital of Guangxi Medical University, including 20 patients with mild pre-eclampsia and 10 patients with severe pre-eclampsia. Twenty normal pregnant women in the third trimester of pregnancy were randomly selected as control group. The expression of VEGF protein in placenta was determined by streptomycin-biotin-peroxidase complex technique. The expression of ELISA in peripheral blood of normal pregnant women and preeclampsia pregnant women was determined by ELISA technique. Results the expression level of XIAPN VEGF in serum of preeclampsia patients was 203.92 鹵19.97 mg / L, respectively. The expression level of XIAPN VEGF in serum of pregnant women in control group was 300.88 + 25.50 mg / L, respectively. The expression of XIAPN VEGF in serum of preeclampsia group was different from that of normal control group (P < 0.05). There was a significant difference between preeclampsia group and normal control group in the expression of XIAPP VEGF in serum of preeclampsia group and normal control group. The expression of VEGF in the serum of mild preeclampsia group was 215.56 鹵9.60 mg / L 122.81 鹵8.24 mg / L respectively. The expression of VEGF in the serum of severe preeclampsia group was 180.62 鹵13.66 mg / L 103.32 鹵5.35 mg / L, respectively. There were significant differences in the relative expression of XIAPP VEGF in serum of severe preeclampsia group. There was a positive correlation between the expression of XIAPP VEGF in serum and the level of XIAP in the control group. The positive expression rates of placenta in mild and severe preeclampsia groups were 100% and 62%, respectively. There was significant difference between the mild preeclampsia group and the control group (蠂 2 / 17.391P 0.000), and between the severe preeclampsia group and the mild preeclampsia group, there was a significant difference between the severe preeclampsia group and the mild preeclampsia group. The positive expression rates of VEGF in placenta of mild and severe preeclampsia groups were 100% and 72%, respectively. The positive rates of VEGF in mild and severe preeclampsia groups were 100% and 72%, respectively. The positive expression rates of VEGF in mild and severe preeclampsia groups were significantly higher than those in control group. The difference was statistically significant (蠂 ~ 2 + 10.497) P ~ (0.001), and there was a significant difference between the severe group and the mild group (蠂 ~ (2 +) 9.269% P ~ (0.002). Conclusion: VEGF may be involved in the development of preeclampsia.
