【摘要】：背景:目前,對(duì)于體外生殖培養(yǎng)液的質(zhì)量控制文件或技術(shù)標(biāo)準(zhǔn)中,對(duì)成分含量的指標(biāo)及檢測(cè)方法未做明確規(guī)定,為保證該類產(chǎn)品的安全有效性,應(yīng)及早建立質(zhì)量標(biāo)準(zhǔn)。目的:建立采用液相質(zhì)譜聯(lián)用法檢測(cè)體外生殖培養(yǎng)液中葡萄糖、乳酸鈉、丙酮酸鈉3種指標(biāo)成分的方法,分析不同用途培養(yǎng)液中3種成分的含量。方法:采用超高效液相色譜三重四級(jí)桿質(zhì)譜聯(lián)用法,使用SUPELCO Discovery HS F5-3色譜柱(15 cm×2.1 mm,3μm),流動(dòng)相為0.1%甲酸水溶液-0.1%甲酸乙腈溶液,梯度洗脫,流速0.35 m L·min-1,柱溫40℃;質(zhì)譜采用電噴霧離子源,負(fù)離子模式掃面,掃描方式為多反應(yīng)監(jiān)測(cè)。結(jié)果與討論:葡萄糖、丙酮酸鈉、乳酸鈉分別在0.1-10 mg/L(r=0.999 8)、0.05-5 mg/L(r=0.999 4)、0.1-10 mg/L(r=0.999 4)范圍線性關(guān)系練好;平均加樣回收率在96.4%-98.1%之間,相對(duì)標(biāo)準(zhǔn)偏差均小于2.8%。說(shuō)明采用液相質(zhì)譜聯(lián)用法檢測(cè)體外生殖培養(yǎng)液靈敏、快速、準(zhǔn)確、專屬性強(qiáng),可為體外生殖培養(yǎng)液的質(zhì)量標(biāo)準(zhǔn)研究提供依據(jù)。
[Abstract]:Background: at present, in the quality control document or technical standard of in vitro reproductive culture medium, there is no clear stipulation about the index of the content of ingredients and the detection method. In order to ensure the safety and effectiveness of this kind of product, the quality standard should be established as soon as possible. Objective: to establish a method for the determination of glucose, sodium lactate and sodium pyruvate in vitro reproductive culture medium by liquid phase mass spectrometry, and to analyze the contents of three components in different culture media.
