玻璃化冷凍胚胎與新鮮胚胎移植對子代小鼠生長發(fā)育的影響

【摘要】:目的輔助生殖技術(shù)會引起子代缺陷,文中通過比較經(jīng)體外受精(invitro fertilization,IVF)的新鮮胚胎與玻璃化冷凍胚胎移植后所產(chǎn)生的子代小鼠生長狀況,以探討胚胎冷凍對IVF后代生長發(fā)育的影響。方法實驗共分為3組,對照組(12只):小鼠自然交配所產(chǎn)生子代;鮮胚移植組(9只):收集IVF后的小鼠囊胚,直接將囊胚進行移植;凍胚移植組(9只):收集IVF后的小鼠囊胚,進行玻璃化冷凍胚胎,復(fù)蘇后移植到假孕母鼠子宮內(nèi),分析比較3組子代小鼠體重的變化,性成熟期體脂。結(jié)果鮮胚移植組與凍胚移植組胚胎在高倍顯微鏡下形態(tài)無明顯差異。凍胚移植組0~12周子代小鼠體重高于對照組(P0.01),鮮胚移植組子代小鼠0周、2~6周體重高于對照組(P0.05);凍胚移植組性成熟期脂肪含量[(9.32±4.34)g]高于鮮胚組[(5.24±2.56)g]和對照組[(2.80±0.48)g],差異有統(tǒng)計學(xué)意義(P0.05)。凍胚移植組肌肉含量[(33.77±4.22)g]高于對照組[(27.50±1.79)g],差異有統(tǒng)計學(xué)意義(P0.01)。結(jié)論鮮胚移植技術(shù)對子代的生長狀況干擾小于玻璃化冷凍技術(shù)。
[Abstract]:Objective to study the effect of embryo freezing on the growth of IVF progeny by comparing the growth status of fresh embryos and vitrified frozen embryos after transplantation of fresh embryos and vitrified frozen embryos. Methods the experiment was divided into three groups: control group (n = 12): offspring produced by natural mating of mice; fresh embryo transplantation group (n = 9): collection of mouse blastocyst after IVF and direct transplantation of blastocyst; frozen embryo transplantation group (n = 9): collection of mouse blastocyst after IVF, Vitrified frozen embryos were transplanted into the womb of female pseudopregnant mice after resuscitation. The changes of body weight and body fat during sexual maturity were analyzed and compared among the 3 groups. Results there was no significant difference in morphology between fresh embryo transfer group and frozen embryo transfer group under high power microscope. The body weight of chilled embryo transplantation group was higher than that of control group at 12 weeks, the body weight of fresh embryo transplantation group was higher than that of control group at 0 weeks, and the body weight of fresh embryo transplantation group was higher than that of control group at 2 weeks, and the fat content of frozen embryo transplantation group was higher than that of fresh embryo group [5.24 鹵2.56g] and control group (2.80 鹵0.48g), the difference was statistically significant (P 0.05). The muscle content of frozen embryo transplantation group [33.77 鹵4.22 g] was higher than that of control group [27.50 鹵1.79 g], and the difference was statistically significant (P 0.01). Conclusion the influence of fresh embryo transplantation on the growth of offspring is less than that of vitrification.
